The Expression And Its Significance Of Matrix Metalloproteinase-2,-9 In Diffuse Axonal Injury

Objective: To study the signification of matrix metalloproteinase-9 , matrix metalloproteinase-2 in diffuse axonal injury (DIA) and the relationship between them in ratsMethods:Make the model of brain diffuse axonal injury in SD rats. 110 well-being adult SD rats with weight of 250-300g was selected in the current research,all rats were raised in condition of optional diets.All rats were divided into 1 control group and 10 injured groups at random.Every group consisted of 10 rats(including control group and injured groups).The 10 injured groups were 0.5hr group,1 hr group,3 hr group,6 hr group,12 hr group,1d group,3d group,7d group,11 d group,14 d group respectively,according to different timepoints. The expressions of MMP-9 and MMP-2 protein at different times were determined by Immunohistochemistry staining, while the neuron apoptosis was investigated by TUNEL,and the relationship between MMP and C-myc,Fas was investigated by SP Immunohistochemistry staining.Results: The expression of MMP -9 and MMP -2 protein turned out to be increased at 3 hours,peaked at 24-72 hour after injury ,while the increased MMP-9 and MMP-2 protein level sustained for 7 days.Then decreased gradually,and returned to baseline at 14 days..They can increase the expression of c-myc and Fas .And the neuron apoptosis were apparently higher in the traumatic group than in the sham-operated group. Compared with the control group.Conclusion: The expression of MMP-9 and MMP-2 protein are induced to increase after brain diffuse axonal injury in rats,it indicates that MMP-9 and MMP-2 protein is probably involved in the neuron apoptosis,thus leading to aggravation of brain injury.
