Objectives:To explore the changes of the nerve cells'apoptosis and the expressions of MMP-9,Caspase-3,Bcl-2 in the rats'hippocampus tissue after diffuse axonal injury and the impact of the matrix metalloproteinase inhibiter on the nerve cells'apoptosis and the expressions of MMP-9,Caspase-3,Bcl-2 in the rats'hippocampus tissue after diffuse axonal injury. To study the protections and its'mechanisms of the matrix metalloproteinase inhibiter for the rats'brain after diffuse axonal injury.Methods:Sprauge-Dawley rats that weighted from 250 to 350 grams were divided randomly into group 1 (normal contrast),group 2(to inject NS into the cerebral ventricle 30 minutes earlier before elaborating the rat's brain—diffuse axonal injured model) and group 3 (to inject matrix metalloproteinase inhibiter-CTT into the cerebral ventricle 30 minutes earlier before elaborating the rat's brain—diffuse axonal injured model). Every group consisted of 10 rats. The rats in group 1 and group 2 suffered from diffuse axonal injury after instant head rotation. All rats were killed at lhour,3hours,6hours, 1day,3days, and 7days after injury(five rats each time) respectively. The nerve cells' apoptosis in the rats' hippocampus tissue was detected by TUNEL and the expressions of MMP-9,Caspase-3,Bcl-2 in the rats'hippocampus tissue were detected by immunohistochemistry staining. The differences among three groups were compared and analyzed.Results:1 There are few numbers of the apoptotic nerve cells in group 1. The numbers of the apoptotic nerve cells in group 2 and group 3 increased significantly (P<0.05). At every time point the numbers of the apoptotic nerve cells in group 3 were significantly less than the numbers in group 2 (P<0.05).2 Compared with group 1, the expressions of MMP-9,Caspase-3,Bcl-2 in group 2 and group 3 increased significantly (P<0.05).3 Compared with group 2, at every time point the expressions of MMP-9,Caspase-3 in group 3 decreased significantly (P<0.05), but the expression of Bcl-2 increased significantly (P<0.05).Conclusions:1 There were the changes of the nerve cells' apoptosis and the expressions of MMP-9,Caspase-3,Bcl-2 in the rats' hippocampus tissue after diffuse axonal injury.2 CTT could inhibit the expression of MMP-9 and the activation of Caspase-3,improve the expression of Bcl-2, enhance the opposition to the apoptosis, reduce the nerve cells'apoptosis.3 CTT could reduce the nerve cells'apoptosis in the rats' hippocampus tissue after diffuse axonal injury. It showed that CTT had some protections for the rats'brain after diffuse axonal injury.
