Objectives: This study aimed to analyse the expression of C-FOS,TGF-β1 and p27 in human middle ear chofesteatomas and to investigate the correlation between its expression and the abilityof erosionof cholesteatomaMethods: Immunohistochemical staining (SP method) of 31 cholesteatomas and 11 external ear canal skin samples fiom patient and 10 external ear canal skin samples fiom healthful man which were taken intraoperatively,was performed for C-FOS,TGF-β1and P27 positivity. The expression of ten were observed under microscope. Then we scanned it into a computer by an image scannerand quantified the gray of them using commercial software.Results: The percent of positive expression of TGF-β 1 and C-FOS in cholesteatoma were 87.1%, 83.9%. Their expression tended to be increased greatly compared with that in the skins of the control groups. P27 positivity was not obtained in cholesteatornas and external ear skin tissues. It showed statistically significant correlation between expression of C-FOS and the ability of erosion of cholesteatoma . There was correlation between the express ion of TGF-β 1 and the ability of erosion of cholesteatoma too.But there was not correlation between the expression of C-FOS and TGF-β1.Conclusion: The expression of C-FOS in cholesteatoma was significally higher compared with that in the skin of extemal auditory of cholesteatoma patients and healthful man . It means that C-FOS play a great important role in the hyperproliferative of cholesteatoma . We can conclude that cytokines play a great rote in the etiology of cholesteatoma too. TGF-β1, growth restrain factor, can restrain the growth of cel and its high proliferation ,make it be a controllable form. P27 positivity was not obtained in cholesteatoma and extemal ear skin tissues, Perhaps it did not play important role in chotesteatoma. If we can decrease the expression of C-FOS in cholesteatoma, and control the produce of cytokines, we may find a new way to treat it.
