Background:Hepatic fibrosis is an intermediate process in chronic hepatic disease, it is also a reversible process, earlier diagnosis and therapy can inverse the process, whereas it will develop to hepatic cirrhosis, which is an irreversible process. The diagnosis of hepatic fibrosis with ultrasonography is mostly depended on the two-dimensional imaging of hepatic parenchyma, although some authors suggested that Doppler flow waveform of hepatic vein could reflect the changes of hepatic parenchyma, this had not widely agreed. In recent years, with the developing of ultrasound contrast agent and it's correlative techniques, new methods had been provided to diagnose hepatic diseases, but up to now, the studies were mainly applied in the diagnosis of hepatic tumor. As we know, with the diffused pathological change occurred in hepatic parenchyma, the correlative hemodynamic change occurred. Contrast agent was a good tracer of blood circulation, overseas authors had performed a primary study of intravenous contrast ultrasound (ICU) in patients with hepatic cirrhosis and fibrosis, and concluded that the change of transit time of contrast agent in hepatic vein may be a sensitive parameter in diagnosis the hepatic fibrosis, it is under confirmation, added to this, are there any other factors will effect the transit time? What is an accurate method to judge the transit time also need further study.Objective:To investigate the feasibility of echo contrast in evaluating the perfusion of normal liver and the diagnosis of the earlier phase of hepatic fibrosis with self-made albumin perfluorocarbon ultrasonic contrast agent, the normal rabbits and fibrosis model rabbits were involved, and investigate the relationship of Doppler flow waveform of hepatic vein and hepatic fibrosis.Methods:There were 10 healthy rabbits and 20 hepatic fibrosis models involved in this study, to conform the success of establishing hepatic fibrosis models, two methods were applied:â‘ 10 rabbits were treated with the thioacetamide (TAA) in the concentration 1.2mg/mL as the onlydrinking water and normal feed to construct hepatic fibrosis models;â‘¡other 10 rabbits were treated with the animal feed which contained 20% grease, and diethyl-stilbestrol were fed with the dose of 10mg every time and twice a week.The liver images of 10 normal rabbits were acquired before and the 1st min after injecting self-made albumin perfluorocarbon ultrasonic contrast agent (UCA) at the condition of Acoustic Density. Then the images were assessed and contrastively analyzed, parameters such as peak intensity (PI), the area under the curve (AUC), the mean transit time (MTT) and the descent half time (HF) of the hepatic parenchyma were measured before and after ICU. To assess the effect of the dose of the UCA to the transit time of the contrast agent in hepatic vein, the compensated time of the appear time of the hepatic vein(CAT),the time to peak of the contrast agent in the hepatic vein(CPT) and the disappear time of the contrast time in the hepatic vein(CDT) were acquired by subtracting the respective basic values.To observe transit time of UCA in hepatic vein in normal rabbits and fibrosis models. Before constructing the models and in the 8th week,the 12th week and the 16th week during the constructing the models, ICU were performed individually, the appear times of the contrast agent in the hepatic artery and in the hepatic vein,the time to peak of the contrast agent in the hepatic vein and the compensated appear time of the contrast agent in the hepatic vein(CAT) and the compensated time to peak of the contrast agent in the hepatic vein(CPT)were observed (the appear time of the contrast agent in the hepatic artery was made as a basis time, and the CAT,CPT were acquired by subtracting the basis time from the appear time of the contrast time in the hepatic vein and the time to peak of the contrast agent in the hepatic vein individually), then those parameters as CAT and CPT acquired before constructing the hepatic fibrosis models and during...
