| Background and purpose : Bladder cancer is the most common tumor of urinary system in our country . The mortality rate of bladder cancer comparing to the other tumor in the body is 8 th in men and 10 th in women . The tendency of incidence of bladder cancer is gradually rising . The pathogenesis of bladder cancer was not definited , but angiogenesis is playing an important role in bladder cancer . The rapid growth of bladder cancer need enough nutriment and take away plenty of metabolize production . Angiogenesis fulfils the demand of bladder cancer . The new vessel is juvenility in polarization . The joint among cells is relax and the wall of vessel is no perfect . All of these make it easy for cancer cells go into the blood circulation and penetrate through the link of vein-lymphatic transfer to around and long distance . Researches indicate that there are affinities among angiogenesis of cancer and some angiogenic factor s , such as VEGF,EGF,FGF . In these factors , VEGF is the most important . HIF-1 can regulate the expression of VEGF and the gene of EPO,Endothelin-1,oxygen regulated proteins and so on . HIF-1 is an important regulated factor to angiogenesis and cellular hypoxia reaction . HIF-1 is composed of two subunits , HIF-1αsubunit and HIF-1βsubunit . HIF-1αsubunit is the functional subunit . In this experiment , through the examination of the expression of HIF-1α subunit we discussed the effect of HIF-1 in bladder cancer and the relation to prognosis(recur and transfer). Methods: The mass of paraffin of bladder transitional cell cancer and part of new samples were divided into four groups : normal group,â… group(pathology gradeâ… class),â…¡group(pathology grade â…¡class),â…¢group(pathology grade â…¢ class) . The expression of HIF-1α and MVC examined by immunohistochemistry and the relationship of the expression of HIF-1α and MVC discussed . The samples of groupâ… ,â…¡,â…¢ were divided into first incidence group and recur group to discuss the relation between HIF-1 and the recur and transfer of bladder transitional cell cancer .Results:1 There was no expressed of HIF-1α in normal group .The expression of HIF-1αwere found in most samples of bladder transitional cell cancer(no matter what group it is).Over expression of MVC was found in â…¢ group than other groups. 2 None MVC was found in normal group . All of the samples of bladder cancer were found the expression of vessel . Over expression of MVC was found in â…¢ group than other groups.3 Lower expression of HIF-1αand MVC in first incidence group compared with recur group .4 No matter comparing among normal group and â… ,â…¡,â…¢ group or between first incidence group and recur group . The tendency of HIF-1α and MVC increasing or decreasing was consistent .Two of them were positive correlation .Conclusion :1 The number of micro vessel was great different in the three groups of bladder transitional cell cancer .More great is the degree of malignant, more great was the MVC .The vessel in the edge of cancer tissue was more than that in the center of cancer tissue . It proves that there are close relation between angiogenesis and growth of cancer tissue .2 Increasing in pathology grade , increasing in the expression of HIF-1α and MVC . Two of them are positive correlation . It was say that there has close relation between HIF-1α and angiogenesis . Angiogenesis was important to soakage and transfer of cancer cell . So HIF-1 has close relation to soakage and transfer of cancer cell .3 There were great difference in the expression of HIF-1 and MVC between first incidence group and recur group . It proves that there are close relation between HIF-1 and the recur of bladder transitional cell cancer . |
