Electroporation is believed to be the transient creation of micropores in lipid bilayer of cell membrane due to the effects of the electric pulses. It is a biophysics process, which can increase the permeation and conductance of cell membrane. This increase allows many molecules, such as aqueous affinity molecules, virosomes, DNA, proteins and dye granules, etc, which can not pass through cell membrane in normal conditions, to go through cell membrane.The electrochemotherapy (ECT) is a technology that uses electric pulse combined with medicament to treat cancer. The foundation of this scheme is to get elect-operation in membrane by electric pulse, sequentially make it easier that medication comes into the cell to bring its effect into play and improve the effect at counteract cancer. Our team chose the S180 sarcoma cells and use the electric pulse produced by the LN-101 Gene Pulser Transfection Apparatus, combined with several anticancer drugs in a series of experiments and got a series results, which provide basis for the feasibility of the ECT.My thesis includes three sections. In the first part, we have studied the electroporation of S180 sarcoma cell by electric pulse. In this paper, S180 sarcoma cell was handled with various voltages, capacity and pulse number. The handled cell was found to be electropored. At the same time, the microvilli was damaged. By thestains of typannan, we have known the influence of electric parameters to the ratio of poration. That is the odds of the electroporation has positive correlation with the voltages and the pulse number and negative correlation with the capacity.The second part was performed for testing the effect of electric pulse combined with adriamycin (ADR) to treat S180 sarcoma of mice. 63 Kunming mice were randomly divided into 4 experimental groups after inoculation of S180 sarcoma: drug group (D+E-) that was only treated with ADR, electric pulse group (D-E+) that was only treated with electric pulse, drug plus electric pulse group (D+E+) that was treated with both ADR and electric pulse,and control group (D-E-) that was without any treatment. The process of drug treatment was to inject 0.2 ml 0.67% (w/v) ADR on sarcoma. Every electric pulse is applied 5 pulses with 600V and 10 μ F. The result was estimated from the growth of the sarcoma, the histochemical observation, the observation from TEM, and so on. The result was that the electroporation could enhance the effect of ADR.In the third part, we have studied the best electric parameters and the best drug time of the electrochemotherapy to cure the S180 sarcoma of mice. The low dose Bleomycin was combined with electric pulse with different electric parameters to treat S180 sarcoma of mice. Put the pulse number to be 5, then set the voltage at 500V, 700V and 900V, and set the capacity to be 6μ F, 10 μ F and 1411 F. There were 9 treat-teams and 1 comparison. The result was the curative effect of the treat-teams were remarkable than the comparison, but there was no obvious difference among the 9 treat-teams. When the electric parameters were 700V and 10 μ F, and the number of electric pulse were 5, 6 7, there was no obvious difference among the 3 treat-teams. Let the drug time be Imin and 15min, the result was that the curative effect of the Imin-team was better than the 15min-team.
