Study On Effect Of Qi Benefiting And Blood Removing Chinese Drugs And Western Medicine On Vascular Endothelial Growth Factor (VEGF) And Transforming Growth Factor Alpha (TGF-α) In Gastric Precancerous Lesion

Objective To study the effect of Gasthic carcinoma-stopping granule on Epidermal'growth factor receptor (EGFR), transforming growth factor alpha (TGF- a ) mRNA and vascular endothelial growth factor (VEGF) in procancerouse lesions of gasthic carcinoma of rats. Methods In animal experiment ,precancerous lesions of gastric carcinoma models were induced by MNNG and placing the spring in pylorus. The rats were divided into treating group, protective group and control group , and treated with Gastric carcinoma-stopping, gastrone and water before the isolation of gastric tissue, respectively. The mucosal sample was taken from antral region of stomach for pathological changs. The immunohistochemical staining was used to the expression of EGFR. The expression of transforming growth factor alpha mRNA and vascular endothelial growth factor mRNA were detected by RT-PCR . Results It was remarkable that the discrepancy of the pathological changs in the control group and the Chinese herbal group , or in the control group and the gastrone group. The level of EGFR was reduced remarkbally(P<0. 05). The RNA indexs (RI) of transforming growth factor alpha and vascular endothelial growth factor expression in control group was higher than the normal , but was decreased after treatment of Gastric carcinoma-stopping granule(P<0. 05). The Chinese herbal group was significantly different from the control group(P<0.05), and not different from the normal(P>0.05). The gastrone group in the gastrone group was not different from the control group,or one in transforming growth factor alpha was different. Conclusion The Gastric carcinoma-stopping granule can prevent and improvethe pathological changes of precancerous lesions gastric carcinoma. The Gastric carcinoma-stopping granule have the effect on the secretory level of the cell gene of, Epidermal growth factor receptor , Epidermal growth factor receptor transforming growth factor alpha mRNA and vascular endothelial growth factor mRNA. . The gastrone can decrease the expression of transforming growth factor alpha mRNA.
