Objective To observe the effects of propofol on cytosolic free Ca2+ in hypoxia cultured myocardial cells of neonatal rats.Methods Myocardial cells from 1-3 days neonatal Wistar rats were cultured in normal condiat ion for 6-8 days, then were divided into 4 groups randomly. Group I wasn't treated, group II, III and IV were bathed with 95%N3+5%CO2 in one direction flow for 40 minutes at 37℃ before 20 minutes reoxygenation for establishing hypoxic-reoxgenation model. Then group III, IV were added into propofol in normal and larger dose respectively. The cell morphology, value of cell survival numbers and cytosolic free Ca2+ were measured in the four groups.Results The cell morphology: In group I and III, the beating rates of cultured neonatal myocardial cells showed unsignificant change, while in group II and IV the rates were slower. Ultrastru-cture examination showed that cells' membrane and orgenells were normal in group I and III, but in group II and IV they were significantly worse. Value of cell survival numbers: Group I and III were more than group II and IV. Cytosolic Free CaJ' : In group II [Ca2+] (781. 64±298. 20nmol/L)were higher significantly than in group I (188. 99 ± 30. 78nmol/L) ( P<0. 05 ) .In group III [Ca2+],(279. 11 ± 106. Hnmol/l) didn' t change significantly comparing with that in group I (P>0. 05) .There was differencebetween in group II and IV (469. 41 + 145. 35nmol/L) (P<0.05).And [Ca2+], in group IV were higher than in group III. Comparing the fluorescence rate value of original and addition 20umol CaCl2 between group II and III, there was difference.Conclusions The modle of hypoxia-reoxyenation successfully brings about the ovei條oad of cytosolic free Ca 2+. The normal dose and larger-dose of propofol can inhibte calcium overload, but the effect in normal dose propofol is better than that in larger-dose. Propofol protects myocardial cells against hypoxia-reoxyenation by inhibit calcium overload. Propofol perhaps can be regarded as an calcium antagonist.
