Objective:To investigate the expression of Ki-67, p27kipl, CyclinD1, CyclinE in Transitional cell Carcinoma of bladder(TCC);To evaluate their relationship with pathological grade.clinical stage and to improve our understanding of the pathogenesis of TCC.Methods:To study the expression of Ki-67, p27kip1, CyclinD1, CyclinE protein in 41 case of paratfin-embedded TCC tissue and in 12 case of cystitis tissues by immunohistochemical method S-P. The positive rate of Ki-67, p27kipl , CyclinD1 , CyclinE were compared with the pathological grade,clinical stage of TCC.Results:1)The mean positive cell rate of Ki-67 was 14.4% in TCC, while it was 0.14±0.04% in cystitis(p<0.05).It in I, II, III of TCC were 11.7±3. 22, 14.3±2.8,16.5±3.6 (p=0. 02) . 2)The positive rate of p27kipl was 39.0%(16/41) in TCC, while it was 100%(12/12 )in cystitis(p<0.01); It in Tis-T1,T2-T4 of TCC were 62.5%(10/16),24%(p<0.05);It in high differentiation and low differentiation of TCC were 44.4%(8/18), 26.1%(6/23) (p>0.05) . p27kipl protein has no significant difference in pathological grade and has negative related with clinical stage.3) The positive rate ofCyclinD1,CyclinE protein were 43.9%(18/41),51.2%(21/41)in TCC, and no positive cell was found in cystitis;CyclinD1 protein in Tis-T1,T2-T4 of TCC were 62.5%(10/16),21.2%(8/25);It in high differentiation and low differentiation of TCC were 55.6%(10/18), 34.8%(8/23). CyclinDl protein has no significant difference in pathological grade and clinical stage (p>0.05). CyclinE protein in Tis-T1,T2-T4 of TCC were 31.3%(5/16), 64.0%(16/25); It in high differentiation and low differentiation of TCC were 22.2%(4/18), 73.91%(17/23); CyclinE protein has significant difference in pathological grade and cilinical stage (p<0.05). 4)Negative expression of p27kipl protein or over-expression of CyclinE protein were significantly associated with high Ki-67 expression (p<0.05). Negative or positive expression of CyclinDl protein was no significantly associated with Ki-67 expression. Conclusion :The results of this study show that the abnormal expression of p27kip1, CyclinD1,CyclinE, Ki-67 play an important role in the genesis and progression of TCC.
