Background:Epidemiological studies have shown that moderate consumption of red wine has more beneficial effects than other alcohol beverages in decreasingthe mobidity and mortality of coronary heart disease.These findings suggest that red wine posses cardioprotective effects.Recently,the further studies in abroad showed that resveratrol,a polyphenol compound existed in red has the antioxidative capacity and free radical scavenging activities,and the protective effects against arrhythmias and mortality induced by I-R.It can also reduce the myocardial infarct size.In the last few years,It has been studying extensively and increasingly.Studies on the cardioprotective effects of resveratrol and their mechanism is a hot-spot topic in the world.We haven't found the reports about the cardioprotective effects of resveratrol in China.Objective:The major objective of the present study was to examine that whether resveratrol,an antioxidant presents in the wine has the protective effects against arrhythmias,myocardial injuries and mortality of rabbits,and the action of pharmacological preconditioning,and the capacity reducing cardiac infarct size. Methods:Anesthetized rabbits were randonly divided into four groups(n=10 in each group):group D(control) was perfused with vehicle(dimethyl-sulfoxide-0.9%NaCl=l:103,v/v,lml/kg).GroupA,B,C were perfused witha bolus of resveratrol(2.5×10-7,2.5×10-6,2.5×10-5g/kg respectively) from a jugular vein 15min before coronary occlusion.The midpoint of left anterior descending branch was occluded by the following procedures:(l) 10min occlusion followed by 30min reperfusion,and (2)4hr occlusion.Observed:(1) The incidence and duration of I-R-inducedarrhythmias(VT,Vf)and mortality of rabbits.(2) The S-T segment elevation(behind point J 0.04 second) induced by I-R in rabbits.(3) The serum GOT,LDH,CPK at the end of 30min reperfusion.(4) The myocardial infarct size at the end of 4hr occlusion. Then,rabbits were preinfused with and without and different dose resveratrol before 15min occlusion, and the incidence of arrhythmias(VT,Vf) and the S-T segment elevation induced by myocardial injuries,and thr value of myocardial enzymes, and. thr myocardial infarct size were compared. RessultS:The incidence and duration of I-R-induced arrhythmias(VT,Vf) and the severity of myocardial injuries(S-T segment elevation,values of myocardial enzymes) and myocardial infarct size were all obvious lower in group C than in group control,(P<0.01). The differences between group B and control were statistically significant. ( p < 0.05 ) .No significant difference was seen at group A and control. (p>0.05) .The mortality induced by I-R in groupB and C was also lower than in group control, (p<0.05).Group A couldn't reduced the mortality induced by I-R in rabbits.The difference of mortality between groupA and control hadn't statistically significant. (p > 0.05 ) .The difference between the difference dose resveratrol was considered to be statistically significant. (p<0.01) .Conclusion:RESV(2.5×10-6-2.5×10-5g/kg) can reduce the incidence and duration of I-R-induced arrhythmias, and the severity of myocardial injuries,and the mortality induced by I-R in rabbits.The resveratrol could induce pharmacological preconditioning,and reduce the myocardial infarct size.
