Objective By observing the effect of swimming on the rat model of the chronic sciatic nerve compression,to investigate whether the moderate active exercise was beneficial to the recovery of the compressed nerve's function and the optimal time of it;meanwhile to study the expression of Ciliary Neurotrophic Factor( CNTF) in the compressed sciatic nerve and the effect of the exercise on that of it.Methods (1)According to the model of chronic nerve compression in the rat designed by Mackinnon ,made 64 models and randomly divided into the model group,the first swimming group and the second swimming group,which there were 32,16,16 respectively .Another 16 models of the sciatic nerve uncompressed but strained were supposed as the blank group. (2)The rats in the blank and model group were caged but lived freely .At the 2 th and 4 th week after compression,the first and second swimming group swam for 2 weeks respectively. (3) To the blank group,determined Sciatic nerve Function Index(SFI) at the 1th ,2th ,3th ,4th week after compression.To the other groups,determined SFI at every week.At the 2th,4th,6th,8th and the 4th,8th and the 6th,8th week after compression,the model group,the first swimming group and the second swimming group determined Motor Nerve Conduction Velocity(MNCV) respectively and took out the compressed nerve to observe the histological changes through the light and electron microscope.At the 4th,6th and the 4th and the 6th week after compression,the above 3 groups measured the expression of CNTF by the immunohistochemical method respectively.Results (1) SFI,MNCV of the blank group: At the 3 th ,4th week,SFI of the blank group was better than that of the model group (P<0.05).But at the 4th week,MNCVof it had no significant difference to that of normal group( P>0.05).(2) Comparison of SFI,MNCV among the model group and the first and second swimming group: at the 4-8th week ,SFI of the first swimming group was better than that of the model and the second swimming group,which there was significant difference among them (P<0.05);SFI of the second swimming group was worse than that of the model and the first swimming group, (P<0.05). At the 4th week ,MNCV of the first swimming group was better than that of the model group( P<0.05),at the 6th week MNCV of the second swimming group was worse than that of the model group ( P<0.05) ;at the 8th week,MNCV of the second swimming group was worse than that of the model and the first swimming group ( P<0.05).(3) The observation of histology :Myelin sheath was the first affected after compression.The changes of its morphology first appeared,then following the changes of the thickness,at last the axon was affected. Demylination appeared at the 4th week .(4) CNTFrNormal nerve had low expression of CNTF and the expression of it synchronized with the function of nerve.Conclusion (1) The moderate active exercise at the early nerve chronic compression could delay the pathological process of the compressed nerve, but at the meta-anaphase exercise should not be carried out. (2)The expression of CNTF synthonized with the function of nerve.
