Primary bronchogenic carcinoma is now a big threat to mankind. There is significant increment of morbidity and mortality of lung cancer around the world since half century before. Among all carcinoma, lung cancer is the first killer to people who live in metropolis in China and is the fourth cause of death in the nation. The strategy to reduce its mortality depends on diagnosis in early stage and perfect treatment.Any anomaly in the signal transduction pathway may lead to transformation in the process of proliferation, apoptosis, chemotaxis and adhesion of cells, which are related with generation and development of lung cancer. Members of transmembrane 4 super family(TM4SF), who were found widely expressed in many tissues of mankind, some of them have been proved to play a role in the process of cellproliferation, signal transduction and chemotaxis. Most of members in TM4SF like CD9 and CD82, were found associated with good prognosis in many cancers with the only exception of CD151, who is associated with poor prognosis in cancers. The expression level and clinical significance of GDI51 in NSCLC has not been well demonstrated yet.Tea polyphenol, which is extraction from leaf of tea, shows particular properties of anti-inflammation, anti-oxidation and anti-tumor. The main components of tea polyphenol are flavanol, chromoco, flavane biethanol and phenic acid. The mechanism of its anti-tumor property is as follow: affectation to signal transduction , gene expression, cell cycle and DNA synthesis of cells as well as inducing tumor cells to apoptosis. Whether the expression level of CD151, which is a member of cell signal transduction, in cancer cells will be affected in vitro by tea polyphenol will be explored in this study. In our study, we analysed GDI51 expression level in samples resected from 38 NSCLC patients and 20 patients with benignant lung nodules by flow cytometry. We also set up in vitro model of tea polyphenol induced decease of tumor cell lines.Objective: Discover the relevance between expression level of CD151 and clinical datum of NSCLC. Discover the relationship between expression level of CD151 and proliferation activity in NSCLC cells. Set up in vitro model of tea polyphenol induced decease of tumor cells. Discover the influence of expression of CD151 in tumor cell lines in vitro by tea polyphenol.Methods: Specimen were obtained from 58 patients who underwent surgery at the Department of Thoracic Surgery of the First Affiliated Hospital of ZheJiang University (HangZhou, China). 38 of which were NSCLC, 20 of which were benignant nodules. It was the first time resection for all patients and there were not any radial or chemical therapy performed before operation. All diagnosis was proved by the Pathology Department in the same hospital. We took 3 specimen from each patients: the nodule, tissues 5cm from the nodule, tissues 10cm from nodule. Tissues were processed mechanically into cell suspension. Expression level of GDI 51 in the tissues was measured by flow cytometry(FCM) after we incubated cell suspension with antibody stained with PE. Also, using FCM, PI and SPF can be measured when we processed cell suspension with PI solution.Set up anti-tumor model by culturing cancer cell lines PG (human pulmonary cancer cell line), KB(human buccal cavity cancer cell line) with different concentration of tea polyphonel. Find out the inhibit effect of cancer cell lines by tea polyphonel with MTT test. Check the expression level of CD151 in cell lines mentioned above after cocultured with tea polyphonel.The data are presented as mean SD and compared with t test and one-way ANOVA analysis using SPSS 10.0 statistical program. Relevance was also analysed with the same program.Results:1. Expression of CD151: expression of CD151 in NSCLC tissues (5.57 2.54)was significantly higher than that in tissues 5cm from the mass (3.44 0.90, p< 0.01), tissues 10cm from the mass (3.35 0.85, p < 0.01) and benignant tissues (3.84 0.92, p<0.05) as well.
