Objective: To investigate the regeneration of peripheral nerves allograft cryopresevated in different temperature and wrapped by auto-extracranial jugular vein.Methods: 40 female SD rats in this experiment were carried out randomly in 4 groups. In group A, a 10cm segments of the transected allogenic peripheral nerve cryopreservated in –196℃ for 2 weeks was transplanted, wrapped by auto-extracranial jugular vein , into the left sciatic nerve. In group B, a 10cm segments of the transected allogenic peripheral nerve cryopreservated in –196℃ for 2 weeks was transplanted, not wrapped by auto-extra cranial jugular vein , into the left sciatic nerve. In group C, a 10 cm segments of the transected allogenic peripheral nerve cryopreservated in 4℃ for 2 weeks was transplanted, wrapped by auto-extracranial jugular vein , into the left sciatic nerve. In group D, a 10 cm segments of the transected allogenic peripheral nerve cryopreservated in 4℃ for 2 weeks was transplanted, not wrapped by auto-extracranial jugular vein , into the left sciatic nerve. 2 weeks later, two segments of transplanted nerves were harvested randomly and examined histologically. 12 weeks later, the regeneration of sciatic nerve was examined with the spread of toes,histology and electro-physiology.Result: In the segments of nerves harvested at 2 postoperative week, the infiltration of lymphocytes in group A was same as in group B; The group C and D had the same infiltration of lymphocytes. But ,the infiltration in group C and D was more serious than in group A and B. At 12th week following operation,from the spread of toes,the electro-physiological and histological study showed that the nerve regeneration of group A was much better than group B and C, while,group D was worse than group B and C.Conclusion: 1. Cryopreservation of graft is beneficial in allograft,better in -196℃ than in 4℃ 2. Wrapping by auto-vein is beneficial in allograft.
