Objective:The reparation and reconstruction of peripheral nerve defect is a problem in the aera of the peripheral nerve.Nerve autograft is the best way of reparing peripheral nerve defect,but it will bring the loss of sensory pf the donor area inevitably and the donor nerve is limited.Many researchers devoted themselves to the study of peripheral nerve and allograft.They tried to improve regenerative effect by lowering the rejection reaction.The ways of lowering the rejection reaction include immunosuppressing the recepter and preserving the donor nerve.Among the ways of preserving the donor nerve,the liqiud nitrogen refrigeration and preservation with glycerin are simpler then the others and there comes better regenration.The purpose of the experiment is to find a way of perpheral nerve allograft by studing and comparing the regenration of the peroheral nerve allograft of siatic nerve preserved with glucerin or liqiud nierogen refrigeration.Methods:40 adult male rats were devided into four groups randomly.There are 10 rats in each group.GroupA(n=10) served as nerve donor.The two side siatic nerve were cut off and 10 of them were preserved under -196 for 3 weeks and they were put into the saline under 37 for 30 minutes before allograft.The other 10 nerves were sequently put into the glycerin for 3 hours each time whose concentration was 50%,70%,85%,then preserved in the glycerin whose concentration was 85% for 6 weeks.The nerves were put into the saline for 30 minutes before allograft.Group B(n=10) was subjected to allograft of siatic nerves preserved with liquid nitrogen refrigeration .Group C(n=10) was subjected to allograft of siatic nerves preserved with glycerin.Group D(n=10) served as control and was subjected to autograft of siatic nerves 0.5cm of the siatic nerve was nerves(deficit 0.5cm),and the two ends of the transevted siatic nerve were seperated lcm(gap 1cm).A 1.2cm siatic nerve graft was used to repair the 1cm gap in the siatic nerve using 11-0 nylon epineurial sutures.Each animals recerived an allograft or an autograft.Each animal received no medicine.The food and the oircunstance of the animals are the samc.The specimens were observed under naked eyes and light microscopes at 4, 8, 16 weeks after operation. Electron microscope, electrophysiological examination and the image analysis of axons were alone at postoperation 16 weeks. The siatic nerves preserved with glycerin or liquid mitrogen refrigeration were observed under light microscopes and electron microscope.ResuIts:The regenerration of group D was best one among the three groups of group C was better than that of group B but was inference to group D.There were significantly statiscal difference in the results of electrophysiological examination and the image analysis of axons(P<0.05).ConcIusioir.The sciatic nerves after being preserved with glycerin can efficiently promote regeneration of peripheral nerve allograft.
