Objective Discuss the irradiating effect of LIM to the pathological process of blood vessel RS and assess the probability of LIM irradiation to prevent RS.Methods 1. Distribute 48 male healthy New Zealand rabbits randomly into 4 groups. Use Fogarty catheter to pull EIA tunica intima establishing RS models. Apply 2450MHz microwave with different power of 2mW/cm2, 5mW/cm2 and 10m W/cm2 locally irradiate EIA (1h/d). Obtain specimens at different time of 3d, 1w, 2w and 4w after operation.2. Observe morphological changes of tissues by HE, EF staining and use image analysis system to analyze the area of tunica intima, tunica media, the rate of cavity stenosis and the cells number of tunica intima; Use TUNEL method to detect apoptosis; TEM method had been used to observe the phenotype and microstructure of VSMC. 3. Use SPSS10.0 statistical analysis.Results 1. After microwave irradiating, inflammatory reaction in early period was suppressed, mural thrombus decreased, the proliferation and migration of VSMC depressed, the area of tunica intima, the rate of cavity stenosis and the cells number of tunica intima obviously reduced, comparing with the control group (p<0.01). 2. The rate of positive cells detected by TUNEL method showed that there were no obviously differences among each group at the time of 3d after operation (p>0.05). At other different time, however, the rate of positive cells in irradiating groups obviously increased higher than the control (p<0.01), particularly in the group with power of 5mW/ c m2 in which it went up respectively by 24.16+2.48%, 50.83+3.87%, 49.50+2.43% at different time of 1w, 2w and 4w after operation. 3. TEM showed that the synthesis form VSMC number in the control occupied 93.50+3.45% of the total number of VSMC at 2w after operation. Most of VSMC appear contractile in irradiating group in which there exist a lot of morphological changes of apoptosis in fibroblast and VSMC.Conclusion 1.LIM irradiation obviously affected pathological process of blood vessel RS. After LIM irradiating, we can see that inflammatory reaction in early period was suppressed, the proliferation and migration of VSMC depressed, LIM irradiation promoted cell apoptosis, effectively prohibited the occurring and development of RS. It has be likely that LIM irradiation would become a effective way to treat and prevent RS. 2.LIM irradiation has had relationship between quantity and effect, power span to effectively prohibit RS, particularly in the group with power of 5mW/cm2.
