Study On The Relationship Between Microsatellite Abnormity At Chromosome 17 And Lung Cncer

Objective: The aim of this study was to investigate the incidence of MSI and LOH of four loci at chromosome 17 in lung cancer, and to analyze the relationship between microsatellite abnormity and clinicopathological parameters of lung cancer.Methods: Four microsatellite loci D17S250,NME1,D17S855 and D17S856 at chromosome 17 were used to examine 39 cases of tumor-normal paired tissues for MSI and LOH using DNA extraction, PCR, denatured polyacrylamide gel electrophoresis and silver-staining methods.Results: 44% (17/39) of the cases showed MSI or LOH at one or more than one locus, and 7 cases (18%) displayed MSI, 14cases (36%) did LOH. 5 cases (13%) exhibited MSI and LOH concurrently. All the 4 cases showed MSI or LOH at different sensitivity. The order was D17S855, (28%,11/39), then D17S856(21%),NME1(10ï¼…),D17S250(8ï¼…). The frequency of MSI or LOH in the early-stage (stagesâ… andâ…¡,53%) was obviously higher than that in the advanced stage (stage â…¢,11%), P<0.05, and the frequency of MSI or LOH in cases without lymph node metastasis (67ï¼…)was higher than that in those with lymph node metastasis(33ï¼…),P<0.05. However, the frequency of MSI or LOH had no statistic difference between high-grade differentiated lung cancer and low-grade ones, P>0.05, Also, no relationship was observed between the presence of MSI or LOH and the histologic type, localization of lung cancer, as well as age or sex of patients, P>0.05.Conclusions: There 's a high frequency of MSI or LOH at some loci of chromosome 17, this presence of microsatellite abnormity could occur early and may play a significant role in the development of lung cancer.
