Purpose :To examine the effects of prevention and therapy of herbal compound RuanJianGanBao on the experimental liver fibrosis(LF) of rats, and the difference expression of TGF β 1 which canonical correction with fibrosis in liver. The mechanism of this therapeutic effect of liver fibrosis will be discussed.Method:Complex factors induced-LF rats were adopted, and dimidiated into preventing and therapy groups. There are 5 teams be partition, normal, model, low dose, high dose and control team in each preventing and therapy group. Serum alanine aminotransferase (ALT), the ratio of albumin and globulin (A/G), hyaluronic acid (HA), laminin (LN), type IV collagen (CIV) were detected. Morphological changes were evaluated by professional pathologist The expression of TGF β 1 during the therapeutic process of liver fibrosis were checked by RT-PCR and western-blot analysis.Results:Serum ALT of all teams given medication decreased markedly (p<0.05) , and the ratio of albumin and globulin (A/G) increased markedly in high dose team .There were marked difference on HA. LN. CIV, between model and high or low dose team of the therapy groups ( p<0.05). Morphological changes: the medication compounds could effectively reduce the degree of liver fibrosis induced by CCL4. It could relieve the degeneration and necrosis of hepatocytes and reduce (all or part) fibrous connective tissue deposited in the liver lobules, approaching to the normal liver tissue basically. Statistics of double blind trial showed, there were obvious difference between high dose and normal team (p<0.01), but no significant difference between the normal and control team (p>0.05) . Expression quantity of TGF β 1 mRNA in normal team differentiate notably from those in the high dose, low dose, normal and control team (p<0.01) by RT-PCR in rat liver. TGF β 1 protein showed marked higher expression quantity in model team by western-blot than normal and all team given medicine (p<0.01). Conclusion:Ruanjianganbao compounds has evidence in preventing and repairing effects on the liver fibrosis induced by complex factors and high dose group is superior than other groups. The mechanism of this therapeutic effect of liver fibrosis correlates tightly with its decreasing the activity of ALT and increasing A/G, decreasing HA, LN, CIV and TGF β 1 expression .
