Total flavone of Abelmischl Manihot L.Medic (TFA) is a flavonol glycoside. The effects of TFA on cerebral ischemia and reperfusion injuries were studied in this paper. In addition, the mechanism of TFA against ischemic cerebral injuries was investigated.1. In miceOn the cerebral ischemia model in mice, 30,60,120mg kg-1 TFA could markedly enhance the survival rate during 6 hours and inhibit the increase of MDA content in ischemic cerebrum. On the nitrogen anoxia model, TFA (30,60,120mg kg-1) could prolong the survive time after anoxia.2. In rabbitsOn the cerebral ischemia-reperfusion model in rabbits, TFA (12, 24, 48mg kg-1) significantly inhibited the decreases of LDH and ATP and increase of MDA in cortex, and promoted the recovery of change of EEG induced by cerebral ischemia and reperfusion.3. In ratsOn the thrombus formation model of artery-veins cross in rats, TFA (25, 50,100mg kg-1) significantly and dose-dependently lessened the weight of thrombus.4. Effects of TFA on platelet coagulation and [Ca2+]i in plateletTFA (0.025, 0.05, 0.1mg ml-1) possessed dose-dependent inhibitory effects onrabbits platelet aggregation induced by collagen. TFA significantly reduced the resting and CaCl2-induced increase of cytoplasmic free calcium concentration ([Ca2+]i) in rabbits platelet in vitro, and the effects were in concentration-dependent manner. 5. Effects of TFA on spinal cord electric current induced by Gly in ratsTFA(0.1, 0.2 mg ml-1) possessed concentration-dependent inhibitory effects on spinal cord electric current induced by Gly in rats, and it is competitive inhibitory effect.Conclusion: TFA had protective effects against cerebral ischemia and,or reperfusion injuries via attenuating cerebral OFR lipid peroxidation, inhibiting the formation of thrombus and calcium influx, improving metabolism of energy substance, and antagonizing the electric current induced by Gly.
