Total flavones in the flowers of abelmoschus manihot L (TFA) is the effective part, which was extracted from a Chinese herb called abelmoschus manihot L. In this paper, the protective effect of TFA on incomplete cerebral ischemia injury and its mechanism were studied. Results were showed as followed:1. 20, 40, 80 mg. kg-1 TFA can significantly and dose-dependtently prolong the time of gasping after cutting head in mice.2. On MCAO (middle cerebral artery occlusion) model, TFA (25,50,100 mg-kg-1,ig) can significantly reduce the weight of cerebral infarction . TFA 100 mg-kg-1 can also inhibit the decrease of the activity of LDH in the cerebrum.3. On MCAO model , the examination of transmission electron microscopy and TUNEL method showed that TFA (25,50,100 mg-kg-1,ig) can dose dependently inhibited the apoptosis of brain cells4. On MCAO model, TFA (25,50,100 mg-kg-1,ig) remarkably increased NO content in the cerebrum in dose-dependent manner.5. On the acute incomplete cerebral ischemia model in rats, TFA (50,100 mg-kg-1,ig) can obviously reduce the water in the cerebrum, and 25,50,100 mg-kg-1 improve the pathology changes of cerebral tissue.6. TFA 15,30 mg kg-1 can increase the cerebral blood flow in dogs, and 30 mg 'kg-1 TFA significantly reduced the peripheral resistance of cerebral vessels, on the other hand ,15,30,60 mg kg-1 TFA has no obvious effects on mean arterialpressure, electrocardiogram (ECG) and heart rate in dogs. As above, TFA play a protective role on incomplete cerebral ischemia injury via inhibition of apoptosis, increase the cerebral NO content , relaxing of cerebral vessel and increasing cerebral blood flow.
