Objective: To Investigate the expression and the significance of endometrial TH1/TH2 cytokmes throughout menstrual cycle and early pregnancy in healthy women and unhealthy women with unexplained recurrent spontaneous abortion (URSA).Method:Forty-three cases of healthy women and forty cases of women with URSA were studied by Immunohisto- chemical method(SABC)using monoclonal antibody to Interleukin-2(IL-2), Interleukin-6(IL-6), Tumor necro-sis factor-alpha (TNF-α ) and In situ hybridization method(ISH) using digoxin-labeled probe to IL-2 messen- ger RNA and IL-10 messenger RNA.Results: 1 IL-2: It was showed that the increasing positive levels of endometrial epithelioglandular IL-2mRNA ,IL-2 throughout menstrual cycle and early pregnancy in unhealthy women with URSA have significant difference among proliferative, secretory, early pregnancy periods (P<0.01).The levels of endometrial epithelioglandular IL-2mRNA, IL-2 of secretory, early pregnancy periods were both significantly higherin unhealthy women with URSA than that of healthy women having a low levels of IL-2mRNA, IL-2(P<0.01).But there were no difference about the expression of IL-2mRNA, IL-2 in proliferative period between the two groups, 2 TNF- α ;The increasing positive levels of endometrial epithelioglandular TNF-α were seen throughout menstrual cycle and early pregnancy in healthy and unhealthy women.there were both significantly statistical difference (P<0.01) among three periods of two groups .but the levels of TNF- a in proliferative, secretory , early pregnancy periods weren't all significantly statistical difference between healthy and unhealthy women (P>0.05). 3 IL-6 IL-10mRNA: in contrary to IL-2,the increasing positive levels of IL-6 ,IL-10mRNA throughout menstrual cycle and early pregnancy were observed and had both significantly statistical difference among proliferative, secretory, early pregnancy periods in healthy women (p<0.01) compared with unhealthy women with URSA having a low levels of IL-6 , IL-10mRNA in three periods(P>0.05). Comparative results of IL-6, IL-lOmRNA in three periods between healthy and unhealthy women show: proliferative: p>0.05,secretory: p<0.01,early pregnancy: p<0.01. Conclusion: TH1 cytokine IL-2 has a immune exclusiverole in human pregnancy and is responsible for URSA. TNF- a may be good to the proliferation , growth, development of endometrium, It may play positive or negative immune roles in human pregnancy by complicated biology characteres. TH2 cytokine IL-6, IL-10mRNA play positive and immune nutritious role in normal pregnany. The results provide the clue that the successful pregnancy is due to the TH1/TH2 cytokines balance in maternal-fetal tolerance.
