Objective:To investigate the effect of IL-1β and Astraglus in expression of cultured airway epithelia cellsMethods: The cultured airway epithelia cells in vivo were randomly divided into treatment group and normal control group, To complete this experiment , we need there steps. (1)Treatment group(3h, 8h, 24h, 36h group)were treated with IL-1β1 ng/ml respectively, normal control group were treated with equal culture medium, supernatant and cell-coated dishes were attained after 3h, 8h, 24h, 36h. (2) Treatment group (IL-1β 0.1 ng, 1ng,10 ng group) were treated with IL-1 β 0.1 ng/ml, 1ng/ml, 10 ng/ml respectively,normal control group were treated withequal culture medium. supernatant and cell-coated dishes were attained after24h. (3) Treatment group(Astraglus 50 mg, 200 mg, 500 mg),IL-1β10 ng/ml and dexamethasone group)were treated with Astraglus 50 mg/ml, 200 mg/ml, 500mg/ml,IL-1β10 ng/ml and dexamethasone100ug/ml respectively, normal control group were treated with equal culture medium. supernatant and cell-coated dishes were attained after24h. The expression of TGF-β, protein was measured by ELISA and immunocytochemistry.Results:(1) The expression of TGF-β1 protein were analyzed after airway epithelia cells were treated with IL-1β(1ng/ml ) at different time (3h, 8h, 24h, 36h). The level of TGF-β1 were significantly higher at 3h, 8h, 24h, 36 h in treatment group than those from normal control group(p<0.05, respectively), but the level of TGF-β1 was significantly higher on the 24h in treatment group than those from 3h, 8h, 36h (p<0.05, respectively) . (2) The level of TGF-β1 were significantly higher in the IL-1β(0.1 ng/ml,1ng/ml,10ng/ml )group than those fromnormal control group (p<0. 05, respectively) . The level of TGF-β1 was significantly higher in the IL-1β (10 ng/ml) group than those from IL-1β(0.1 ng/ml andl ng/ml)group (p<0.05, respectively) . At IL-1β concentration ranging frorn0.1 ng/ml ,1 ng/ml,10 ng/ml, a positive dose-effects correlation was found between TGF-β1 secretion and IL-1 β concentration(r=0.906, P<0.01). (3) the concentration of TGF-β1 from Astraglus 50mg, 200mg, 500mg group and dexamethasone group were much more lower than IL-1β (10 ng/ml) group (p<0. 05, respectively) , the concentration of TGF-β1 from Astraglus 50mg, 500mg group were significantly higher than Astraglus 200mg group (p<0.05,respectively), but the concentration of TGF-β1 from Astraglus 200mg group was not different from dexamethasone group (p>0. 05) .Conclusions: It was suggested that IL-1β might accelerate airway remodeling occur and develop by decreasing the TGF-β1 expression and that Astraglus might prevent airway remodeling by increasing the TGF-β1expression,which may contribute to its antiinflammatory effect.
