Objective To research the expression of Platelet-derived growth factor-BB(PDGF-BB)and interleukin-1β(IL-1β)in the rat airway remodeling models of asthma, and to study the relationship among the expression of PDGF-BB.IL-1βand the airway remodeling. The effects with dexamethasone on them were also observed.Methods Forty wistar rats were randomly divided into 4 groups : control group(C group);asthma group(N group);earty–interventional group(E group);late-inerventional group(L group). Each group contained ten rats.Each rats which were N group,E group and L group used intraperitoneally injection with 10% ovalbumin antigen suspension to sensitize on the first and eighth days in the study and began from the fifteenth day ultrasonic atomizing inhalation with 1% ovalbumin eight weeks and each day 20min to stimulate asthma ,to establish the asthma model.Began from the first day and fifteeth day of ultrasonic atomizing inhalation,each rat in the E group and L group intraperitoneally injected into dexamethasone(0.5mg/kg) one hour before each ultrosonic atomizing inhalation .C group was replaced by sodium chloride.Lung tissue were sliced and hematoxylin and eosin stained.The airway morphological parameters was measured by computer image analysis system to estimate the degree of airway remodeling.Expression of PDGF-BB was measured by immunohistochemical method. The expression of IL-1βin blood serum were measured by ELISA method. The data was analysed by SPSS 16.0.Results The airway remodeling were observed in various groups , which include E group, L group and N group. The airway remodeling degree( thicknesses of bronchus wall and airway smooth muscle )in E group were significantly thinner than that in L group and N group (P<0.05), the expression of PDGF-BB and IL-1βin E group were lower than that of L group and N group (P<0.01), and the expression of both PDGF-BB and IL-1βwere related with the airway remodeling degree( thickness of bronchus wall and the thickness of airway smooth muscle) in asthmatic rats(P<0.001).Conclusions PDGF-BB and IL-1βwere related with airway remodeling in asthmatic airway remodeling models of rats,Early treatment with dexamethasone could partly reverse the airway remodeling of asthma, and the expressions of PDGF-BB and IL-1βmay be inhibited . Which suggested that they might become one of indicators to evaluate the extent of airway remodeling.
