Mumps, Tonsillitis are clinic ordinary diseases. The purpose of this project is to research a safety, effective medicament that can prevent and cure such diseases.Puqin Xiaoyan Tablets is made up of Radix Sculellariac , Herba Taraxaci Fructus ForsythiaeRadix Isatidisctc. and has the actions of clearing heat and expelling miasma, anti-inflammatory effect and subsidence of swelling. It is prepared to be tablets by prescription filtration prescription argumentation based on the clinical soup dosage. This project mainly deals with the prcparation procedure, qualily standard and stability of the prcpration.The optimum extraction process of Radix Scutcllariac was investigated by L (3) orthogonal experimental design as the baicalin content index. The optimum extraction process was as follows: Radix Scutcl lariac was extraclcd with 60% alcohol for 3 times, with the solvent volume 8 times amount and extraction time 1.5, 1.5, 1h in turn.The optimum extraction process of Fructus Forsythiac and other five drugs was selected with the L (3) orthogonal experimental design as the forsythin content index. The optimum extraction process was as follows: adding 11 limes amount of water for the first time and 8 limes for the second lime and the third lime, and extracting for 2 hours each time.The method of fluid bed granulation can simplify the procedure and reduce the amount of excipicnts. To select the optimum conditions for fluid bed granulation, orthogonal experimental design L(3) was used. The results of the study were that the rate of spraying and the blast temperature were notable effect. The optimum process: the rate of spraying was 25g/min, the spraying press was 2.1kg, the blast temperature was 100±2℃ and the aeration temperature was 40±2℃.To select the optimum adjuvants for the formulation process of Puqin Xiaoyan Tablets, Starch, Carboxymehtyl starch sodium, Microcrystalline cellulose, Lactose and Starch 1500 were investigated as the hardness, breakage, the disintegration time indexes. It was feasible that the combined excipicnts of Starch , Microcryslalline cellulose, Carboxymehtyl starch sodium (in a rate of 5:5:2) were mixed with granulation. Moreover, the price of such three adjuvants was lower.In the quality control study, Thin Layer Chromatography was used for identification of Herba Taraxaci, llerba Corydalis bungeanae, Herba Bupleuri and Radix Scrophulariae. These TLC methods were simple, rapid and with a good reappearance, and available for quality control of the preparation. The content of baicalin and forsythin in Puqin Xiaoyan Tablets were determined by HPLC. It was limited that the content of baicalin wasn't lower than 8.0mg per tablet and the content of forsythin wasn't lower than 190ug per tablet.The stability of the preparation was investigated. The result wasthat it was stable.
