Immunohistochemical Detect Of TARC And CCR4 In Lesional Skin Of Patients With Bullous Pemphigoid

IntroductionChemokines are small secreted proteins secreted by many cells. They play an important role in regulating migration of leukocytes. Thymus and activation -regulated chemokine (TARC) , a CC chemokine, was identified as a selective chemoattractant for CC chemokine receptor 4 (CCR4) - expressing cells to ti-uss.Bullous pemphigoid ( BP) is an autoimmune subepidermal blistering skin desease, considered to be a Th2 - dominant disease, Th2 type cytokines, IL -4,IL -5 and IL - 13, exist in lesional skin of patients with BP. It was reported that expression of TARC was strongly upregulated in epidermal kerationcytes and langerhans cells by stimulating with some cytokines. We examined the expression of TARC and CCR4 in lesional skin of BP by immunohistochemical procedures.Material and methods1. 12 cases of skin biopsies from patients with BP, 12 biopsies from pemphigus patients, 5 cases of health skins were examined respectively.2. immunoperoxidase labeling was performed using standard technique on 5 m sections. Briefly the frozen tissue were air-dried, fixed in actone, and washed in phosphate buffered saline (PBS). Following protein block with 0. 3% H202, biotinylated rabbit anti-goat IgG, ABC Elite tertiary comples , and AEC solution were added in sequence with intervening PBS washes.3. Observing index : We observed the intensity of immunoperoxidase labe-ling , the positive cells are keratinocytes and infiltrating cells which are stained with orange-red.Resultes1. Strong immunoreactivty of TARC was observed in the epidermal keratinocytes of 11 cases of the lesional skin of patients with BP. One cases of pemphigus was observed expressing TARC in keratinocytes. No immunoreactivity of TARC was detected in normal skin.2.11 cases of BP skin lesions were observed expressing CCR4 +cell in keratinocytes and infiltrating cells. One case was observed in lesional skin of patients with pemphigus. No immunoreactivety for CCR4 + cells was detected in normal skin.ConclusionTARC and CCR4 expression were strongly upregulated in epidermis of BP skin lesions; and they were almost not expressed in pemphigus skin lesions; and they were not expressed in normal skin.CCR4 was strongly expressed on invading cells in dermis of BP skin lesions; and CCR4 and TARC were almost not expressed in pemphigus skin lesions; and they were not expressed in normal dermis.In summary, we showed TARC was mainly secreted in epidermis of BP skin lesions, and attracted the CCR4 + Th2 cells trafficing to skin tissue, and changed the microenvironment of BP skin lesions.
