The research indicates the major reason of tumor occurrence is abnormal cell apotosis. Anti - cancer drugs were applied by causing apotosis to inhibite and clear tumor cells while normal cells were aslo caused apotosis at the same time. Therefore , enhanced apotosis was the reason of chemical therapy side -effects. This adopted ovario - cancer patients as subjects, formation changes of apotosis cells as basis to detect peripherally white blood cell and analyze the relationship of the decreasing and the apotosis of white blood cell and discuss the possible that if the early decreasing of white cell could be predicted by cell apotosis. We try to apply the apotosis of white blood cell to clinical as early diagnose .Method25 ovira - cancer patients who accepted operation and chemical treatment. CP method was applied. We collected vein blood 2ml respectively before chemical treatment in 24 hours and after chemical treatment 5 days. Using hepatin an-ticoagulation: detetced blood routin, cellaptosis respectively by fluorescentmi-croscope and transmission electron microscopy and FCM.1. Count of white cell: detected total number of white cell before chemical treatment and after chemical treatment 5 days respectively.2. Detection of peripherally white blood cellQuantity analysis of peripherally white blood cell with fluorescentmicro-scope, collect coagulated blood and centrifuged to make white cell (1x107个 ml) , observed with fluorescentmicroscope and record datum to analyze.Quantify analysis of apotosis cell with transmission electron microscopy and photography and analysis.Quality analysis apotosis nucleus by FCMAnalyzed datura with FIT cell circle analysis soft system 2.0( Bection Dickinson).Statistic analyzationAdopted with T test and analyze correlation between declining of white blood cell and increasing of apotosis cell.Result1. The change of total number of peripheral WBCThe WBC number of patient decreased significently after chemical treatment , The number of peripheral WBC was 4.1-8. 6xl09 and 2.0 -7. 9 x 109re-spectively before and after chemical treatment.2. Quantity analyze of white cell apotosis with fluorescentmicroscope Compared with that before chemical treatment , the percent of apotosis cellafter chemical treatment increased significently.Correlation of total number of white cell and apotosis cell is obviously , R = -0.736(P<0.05).3. Detect cell apotosis with transmission electron microscopy4. Detect peripheral WBC apotosis with FCMThe percent of WBC apotosis after chemical treatment increased significently compared with that before chemical treatment; correlation analysis with FCM, the increasing of total number of peripheral WBC is related with the number of cell apotosis , R = 0. 646 ( p < 0. 05 ) ; the sensitivity of cell apotosis as early diagnose method of declining of WBC by clinical epdemic analyzation; The sensetivity is 82% , specialiaty is 75% , accuration is 80% .DicussionCytotoxic is major reason of marrow inhibitation caused by anti - tumor drugs. The death of mature granulocyte represent the decreasing of WBC caused by drugs. Generating and cleaning of hemecybe is controlled by apotosis in blood system. Granulocyte exits in blood about 12 hours after maturing, and sc-me vanish on lung and gastro intestinal tract surface. If there is not inflammatory reactive center, apotosis would happen and granulotosis of peripheral blood cell is the prerequisition, that is , apotosis happens before the vanish of WBC in circle system. The determination of apotosis may help to predict the declining of WBC.The chemical treatment of ovario - cancer is long and most of anti - cancer drugs are limited due to their side - effects, while the drug dose is an important factor determining the drug efficiency once granulocyte decreased severely led by narrow transplant, patients were threatened by bacterium and fungi.
