Objective: To study the growth of cell electroporation with low concentration cisplatin affect the ovario-cancer SKOV3 in vitro and the effect of cell elctroporation used alone and in combination low concentration cisplatin in the growth of human ovarian cancer transplanted subcutaneously in nude mice. Methods: 1 , The electric field 600V/cm , electroporation impulse 5 and capacitance 11 μ F were selected to act on ovario-cancer SKOV3-cell line, then according to different action divided into 4 groups: E+M groups, E groups, M groups and contral (C groups). The cell growth after the different treatment and the count of apoptosis condition were observed by flow cytometry, 2 , Creation the model of transplanted tumor subcutanous , randomly divided into four groups in the first experement and treated as follow: (1) E+M group: injected 0.2ml cisplatin (2 μ g/ml) inthe local tumor and electroporation of the tumor , (2).E group: injected 0.2ml 0.9%NS and the electroporated, (3). M group : injected 0.2ml cisplatin (2 μ g/ml) in the tumor. (4). Group C: injected 0.9%NS 0.2ml in the tumor .The different treatments were served from 4 weeks after transplanted and all mice were sacrificed after 9 weeks .The tumor volume and weight, blood CA125, VEGF mRNA and VEGF KDR in the tumors tissue, apoptosis of the tumor cell were detected with the methods were immunohistochemistry, RT-PCR and TUNEL. Result: 1. The result was 2 μ g/ml of the cisplatin in M groups weak growth suppression effection on the ovario-cancer SKOV3-cell line. The group of pure electroporation (E) will make stronger suppression effection than the group M, there were satisfactory suppression effection on the ovario-cancer SKOV3-cell line in the group of E+M and significant difference with E, M and C groups by Statistics analysis (P<0.01) . 2. The apoptosis of E+M group increased (the most at 17.2%, and E,M, C respectively was 4.8%,4.3%,4.1%)the difference was significant(P<0.01) .Advanced Changes of the cell cycles diplayed that, apoptosis of E+M was mainly changed in the G2 -period and in the S -period after the treatments of 3 times into blocked. 3. Body weight of the every group nude mouse had not changed after six times treatments. The comparison of the volum of the different tumor ,the volum decreased significantly of E+M group ,the rate of suppression of E+M group was82.21% to the most, the difference was significant with En M and C groups (PO.01) . 4. The sem-quantu of the VEGF mRNA of E+M was significantly decreased in comparison with E> M and C groups (P<0.01) . 5. After therapies, the CAi25 value in blood in E+M group (7.68 + 1.91) was significantly decreased in comparison with others. 6. Detected the expression of VEGF receptor KDR with the technique of Immunehistochemistry .The result was apparently decreased in E+M group, the KDR was signifficantly differernt with En M and C groups (PO.05). 7. By the technique of TUNEL, the exponent of apoptosis in the tumor cell in E+M group was significant difference with the E> M and C group (PO.05) . Conclusion: 1. It could be the paramete to treat the SKOVrcell line in vitro and animal model when the electrical field is 600V/cm, the number of electroporation is 5 and the capacitance is 11 u F. 2. Electroporation with low concentration cisplatin could suppress obviously the growth of the SKOV3-cell line in vitro and animal model. 3. The electroporation could enlarge obviously the effect of drug resistance of cisplatin with the dosage of non-cell- depress, the method was safety and reliable.
