Objective To study the aromatase mRNA expression in subcutaneous and omental adipose tissues of postmenopausal females and their relation with age ,body weight index , waist to hip ratio , blood pressure,total cholesterol, serum triglyceride and fasting plasma glucose and fasting plasma insulin .Methods Subcutaneous and omental adipose tissues were collected from 30 postmenopausal women undergoing abdominal surgery. Tissues were immediately froze in liquid nitrogen and kept at -70 C for further RNA extraction. Patients, aged from 54 to 65 years, included 10 obesity, 10 obesity with endometrium carcinoma , 10 subjects of normal body weight women. The expression level of aromatase mRNA in adipose tissues was assayed by Semi-quantitative reverse transcriptase polymerase chain reaction amplification. We measured total cholesterol, serum triglyceride and fasting plasma glucose, fasting plasma insulin. The datas assessed by analysis of variance, paired-samples t test and q test, correlations of multiple variables by multiple stepwise regression analysis.Results In postmenopausal women , the aromatase mRNA expression in subcutaneous was significantly higher than that in the omental adipose tissues (Obesity groups: 0.842 + 0.083 VS 0.398 + 0.022 P<0. 001;Controls: 0. 398 + 0. 022 VS 0. 345 + 0. 032 P<0. 001). The level of aromatase mRNA in obesity group was significantly higher than that in controls at both sites(SC : 0. 842 + 0. 083 VS 0.715+0. 054 P<0. 001;OM: 0. 398 + 0. 022 VS 0. 345 + 0. 032 P<0. 001). In subdivision groups, no differention of aromatase mRNA expression in adipose tissues was found between obesity and obesity with endometrium carcinoma groups (SC: 0. 839 + 0. 084 VS 0. 845 + 0. 083 P>0. 05; OM: 0. 398 + 0. 027 VS 0. 397 + 0. 016 P>0. 05). Aromantase mRNA expression levels of subcutaneousand omental adipose tissues in the obesity were positively correlated with waist to hip ratio and fasting plasma insulin (SC: p=0. 019;p=0. 040, OM: p=0. 031; p=0. 022) .Conclusion In postmenopausal , the expression level of aromatase mRNA in abdominal adipose tissues in obesity group was significantly higher than that in normal body weight women, aromantase mRNA expression in adipose tissues was positively correlated with waist to hip ratio and hyperinsuinism.
