Rutoside (Ru) is a flavonoid glycoside,which can be extracted from many herbs. Several study showed that Ru has many pharmacological activity, such as anti-inflamm-ation,anti-virus,reduction of blood capillary fragility and so on. Now it hasn't been reported about the effect of Ru on the disturbance of pancreatic microcirculation in acute pancreatitis(AP).The purpose of this study is to investigate the effect and possible molecular mechanisms of Ru on the disturbance of pancreatic microcirculation of experimental AP induced by sodium taurocholate (STC) in rats. The main contents are as follows:1. Protective effect of Ru (sc) on STC AP in ratsAP model was induced by retrograde injection of sodium taurocholate into biliopan-creatic duct in rat. Ru (30,60,120mg / kg) were administered subcutaneously before the induction of AP. Prophylactic Ru significantly inhibited the increase of serum amylase (AMS) activity, obviously decrease ascites volume and pancreas wet weight. The pathological examination results showed that pancreatic edema, necrosis, inflammation and vacuolization in AP rats at 12h after the induction was effectively suppressed by Ru (sc).The actions indicated a dose-dependent manner.2. Effect of Ru (sc) on disturbance of pancreatical microcirculation in experimental AP The result of hemorheological tests showed that three doses of Ru could decreasewhole blood viscosity, plasma viscosity, erythrocyte sedimentation rate, hematocrit, erythrocyte agglutinatin index and rigidity index levels of AP rats. Ru (60,120mg / kg) could remarkdly reduce pancreatic capillary permeability of pancreas tissue. Meanwhile, it could obviously increase the pancreatic blood flow and improve pancreatic microcirculation.3. Possible mechanisms of improvement of Ru on the disturbance of pancreaticalmicrocirculation in APThe molecular mechanisms of improvement of Ru on the disturbance of microcirculation in acute pancreatitis might related to inhibite the activity of phospholipaseA2 (PLA2), decrease nitric oxide (NO) level, strengthen endogenous antioxidative system and the ability of scavenge oxygen-derived free radicals (OFR) and regulate the balance between thromboxane 82 (TXB2) and prostacyclin (PGF1a).
