From the roots and rhizomes of two folk medicines: Diphylleia sinensis Li., Sinopodophyllum emodi. (Wall.) YING, some ingredients were separated and identified by using modern technologies. At the same time, pharmacology research about one new compound (compound 3) was also investigated in this paper. The main contents and conclusions are provided as follows:1. Seven compounds were separated and identified from the roots and rhizomes of Diphylleia sinensis Li.. They are respectively as followings: kaempferol, Podophyllotoxin, diphyllin, 4-demethyl-podophyllotoxin, picropodophyllotoxin -glucopyranoside, and two unknown compounds.2. Based on previous work, one new aryltetralin-type lignan glycoside was identified by means of spectral data (NMR, MS, CD) : L-4, 5-didemethyl-podophyllotoxin 7' -O-B-D-glucopyranoside.3. The in vitro cytotoxicity of compound 3 (4-demethyl-picropodophyllotoxin 7' -O-B-D-glucopyranoside) was studied on human tumor cell lines (Hela, SH-SY5Y, S180, K562, A549, CNE). Result shovrsobvious cytotoxicity on tumor cell lines, and the cytotoxicity is doseand time dependant. Cell cycle was analyzed by flow cytometry, and theresults indicated that it blocked tumor cells in G2/M phase.
