Objective Carcinoma of large intestine is one of the common malignant carcinomas. Its morbidity and mortality are just less than carcinonas' of stomach and esophagus in the malignancies of digestive system. Currently, it is difficult to diagnose and its 5-year survival rate after operation is low. It threatens health of people greatly. The thesis studied the expression of mdm2, P53, cyclin D15 P16 to explore their roles in the ontogenesis of carcinoma of large intestine.Methods mdm2, P53, cyclin D1, P16 gene protein of 63 carcinomas of large intestine (31 men and 32 women, mean age 56+10 years) and 11 normal large intestine tissue specimen (6 men and 5 women, mean age 55 + 9 years) were measured using flow cytometrytechnique (FCM). The expression of protein was expressed by fluorescent index (FI). The samples in malignancy group were compared according to their clinical pathologic stage, degree of differentiation of carcinoma cell,and histological type. All the results were analyzed with the statistic softwere of ssps8.0.Results (1) The FI of mdm2, P53, cyclin Dl5 P16 (carcinoma of large intestine versus normal mucosae) were respectively 5.96+0.31 vs.3.98 + 0.28 ,P<0.001; 3.79 + 0.52 vs. 4.21 +0.34,P<0.02; 5.44 + 0.39 vs.3.63 + 0.33,P<0.01; 4.21 + 0.89 vs. 4.94 + 0.65,P<0.02. Proliferation indices (PI) of carcinoma of large intestine and normal mucosae were 27.65 +7.96vs. 15.93 + 6.51, P<0.001. (2) Duke's stage C+D versus Duke's stage A+B: mdm2: 5.97 + 0.29vs.5.33 + 0.27,P<0.01; P53: 3.05 + 0.43 vs.3.78 + 0.35,P<0.01; cyclinD,: 5.72 + 0.44 vs.5.11 + 0.31,P<0.01; P16: 3.67 + 0.72 vs. 4.11+0.57 P<0.01. (3) The FI of mdm2, P53, cyclin D1, P16 (infiltration and ulceration type versusprotrusion type ) were respectively 5.74 + 0.43 vs. 5.21+0.31; 3.10 + 0.43 vs. 3.52 + 0.51; 5.76 + 0.43 vs. 5.14 + 0.32; 3.60 + 0.79 vs. 4.14 + 0.68; All P<0.01. (4) The differences were significant between the low differentiation adenocarcinoma and high differentiation adenocarcinomagroups in FI of genes(mdm2: 5.86+0.51vs.5.44+0.41, P<0.01; P53: 3.15+0.64vs.3.92+0.43, P<0.05; cyclinD1 : 5.77+0.41vs.5.43+0.37, P<0.01 ; .P16: 3.82+0.69vs.4.40+0.45,P<0.01. (5) There were no dilferences of FI of genes between PI <27.10 group and PI>7.10 group,P>.05.Conclusions (1) The occurrence of carcinoma of large intestine is related to high expression of mdm2 and cyclin D1. (2) The occurrence of carcinoma of large intestine is related to low expression of P53 and P16. (3) mdm2 and cyclin D1 are related to genesis, infiltration and metastasis of carcinoma of large intestine, while P53 and P16 play negative regulation role. (4) There is no relation between proliferation indices and the expression of gene protein. According to our study, the mdm2, P53, cyclin D1 and P16 gene may act as important indices of clinical diagnosis, treatment and prognosis.
