ObjectiveGliomas are the most common primary brain tumors,which accounted for about in the central nervous system tumors.The prognosis of patients with malignant gliomas is very poor.lt is difficult to eradicate by operation because glioma shows invasive growth.There are high recurrence rate after operation. The purpose of this study is to investigate expressions of MDM2and p21WAF1 proteins in different grades gliomas and their biological significance in tumorigenesis and progression of human gliomas.MethodsThe paraffin-embedded specimens of gliomas were taken from department of neurosurgery in First Affiliated Hospital of Zhengzhou University from June 2007 to July2009,including 58 males,38 females, mean age was 23 ages to 65 ages, the averae age is 42 years old. Pathological grading was classified according to the standard pathological grades for glioma. GradeⅠ19 cases,gradeⅡ31 cases,gradeⅢ22 cases,gradeⅣ24 cases. We combined gradeⅠand gradeⅡof glioma to low grade glioma(LGG),and gradeⅢand gradeⅣto high grade glioma(HGG).The MDM2 and p21WAF1 proteins were determined in the normal,the LGG and the HGG by imunohistochemical Streptavidin- peroxidase (S-P)technique. We respectively use x2 test and Fisher exact probability method to calculate the difference between the normal brain tissue and gliomas,then use Spearman rank correlation analysis to analyze the relevance of MDM2 and P21WAF1.Results1.MDM2:The positive rates of MDM2 protein were 26.00%(13/50) and54.35%(25/46) in the LGG and the HGG (χ2= 8.051, P=0.005). The positive rates of MDM2 increased with the malignant degree.there was a positive correlation in comparison between the expressions of MDM2 and the grades of gliomas by Spearman correlationanalysis (rs=0.290,p=0.004).2. p21WAF1:The positive rates of p21WAF1protein were 74.00%(37/50) and 41.30%(19/46) in the LGG and the HGG (χ2= 10.537, P=0.001). There was a negative correlation between the p21WAF1 expression and the grades of gliomas by Spearman correlation analysis (rs=-0.331, P=0.001).3. There was significant deviation between MDM2 and p21WAF1,there was dependability between MDM2 and p21WAF1(rs=-0.359,P=0.009).Conclusions1.The MDM2 and p21WAF1 play different role in glioma,.the forme rmay function as carcinoma carcinostetic gene in the development of glioma,whereas the later acted as a part in carcinostetic gene.2. MDM2 in normal brain tissue did not express,and the expression of MDM2 was positively correlated with the glioma malignancy.3. p21WAF1 in normal brain tissue did not express,and the expression of p21WAF1 was negative correlated with the glioma malignancy.4.There was significant dependability between MDM2 and p21WAF1. They play a synergistic role in the invasive growth of glioma cells.They are helpful to judge the malignancy,invasion and prognosis of glioma,and become new directions in targeted therapy of glioma.
