Objectives: In view of the fact that lung cancer still represent an important cause of tumor death.Its high metastasis and easy to recurrence after surgery are the fact that cause the low survival of the patients with lung cancer.So efforts should be spent to improve early diagnosis and to identify prognostic factors and to monitor the response of the treatments.The aim of this study is to discuss relationship between serum MMP-2 ,TPA and metastasis,recurrence and biological behavior of the lung cancer so that we can find a prognostic indicator of lung cancer to guide the treatments and monitor the response of the treatments to improve the survival of the lung cancer patients.Methods: Serum MMP-2,TPA levels were detected in 58 patients with lung cancer,15 patients with pulmonary benign diseases and 5 healthy volunteers by enzyme linked imunoassays(ELISA).In lung cancer patients,we observed the relationship between serum MMP-2,TPA and histotype,clinic stage,sex and age.Comparing serum MMP-2,TPA levels with the cases with lymph node metastasis and the cases with non-lymph node metastasis, Comparing serum MMP-2,TPA levels with patients undergoing surgery.In the mean time,measuring the serum MMP-2,TPA levels in the patients pre-themotherapy and post-themotherapy.Results: Mean level of MMP-2 was significantly elevated in cancer samples compared with benign diseases and healthy wolunteers(330.547±164.317ng/ml vs 122.046±43.731ng/ml,50.060±10.865ng/ml,P<0.01).the level of MMP-2 was a little higher in SCLC than that of in NSCLC (368.763±192.685ng/ml vs 315.988±152.205ng/ml).The level of MMP-2 was higher in adenocarcinoma than that of in squamous cell carcinoma(368.529±152.755ng/ml vs 280.260±143.996ng/ml),however these differences were not statistical significance(P>0.05).Serum MMP-2 level was significantly elevated with clinic stage(P<0.05).MMP-2 level in the cases with lymph node metastasis was higher than that of in non-lymph node metastasis(374.439±156.774ng/ml vs 204.720±115.386ng/ml,P<0.01).22 patients undergoing surgery ,the MMP-2 level was higher in recurrence cases than that of in non-recurrence cases(281.350±60.907ng/ml vs 185.044±90.478ng/ml, P<0.05).10 patients who recepted chemotherapy,the MMP-2 level was significantly decreased with treatment(P<0.01). Mean level of TPA is significantly elevated in cancer samples compared with benign diseases and healthy volunteers(4.109±2.770ng/ml vs 1.434±0.379ng/ml,1.425±0.312ng/ml,P<0.01). the level of TPA was higher in SCLC than that of in NSCLC (5.319±3.309ng/ml vs 3.648±2.424ng/ml,P<0.05).The level of MMP-2 was higher in adenocarcinoma than that of in squamous cell carcinoma(4.107±2.408ng/ml vs 3.336±2.433ng/ml),but this difference was not statistical significance(P>0.05). Serum TPA level was significantly elevated with clinic stage.TPA level was significantly higher in stage IV than that of in others stage(P<0.01),but this difference was not significance between stage I and stage II(P>0.05). TPA level in the cases with lymph node metastasis was higher than that of in non-lymph node metastasis(4.819±2.855ng/ml vs 2.073±0.895 ng/ml,P<0.01).22 patients undergoing surgery ,the TPA level was higher in recurrence cases than that of in non-recurrence cases(3.217±0.992ng/ml vs 1.795±0.731ng/ml, P<0.05).10 patients who recepted chemotherapy,the TPA level was significantly decreased with treatment(P<0.01).The serum MMP-2 level was related to the Serum TPA level(r=0.517,P<0.01).Conclusions: ①Serum MMP-2 and TPA levels are significantly higher in lung cancer patients than these of in benign patients and healthy volunteers.This proves that the relation of MMP-2,TPA and lung cancer origination and development .②Senum MMP-2 and TPA levels are correlated with extant of the disease ,and the levels of MMP-2 and TPA are elevated with more advanced stages .③ Serum MMP-2 and TPA levels are relation with metastasis and recurrence.The higher levels of MMP-2 and TPA predict a poor prognosis.④Serum MMP-2 and TPA levels can response the...
