Purpose: To investigate the expression of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in aqueous humor of patients with glaucoma. To initial reveal the internal association between the expression of MMP-2 and TIMP-2 and several types of glaucoma. To further illuminate the associativity with disequilibrium of MMP-2 and TIMP-2 between pathogenesis of primary open-angle glaucoma(POAG). To provide experimental data and theoretical knowledge for the early diagnosis and treatment of POAG.Methods: All objects were divided into five groups. There were 14 eyes of 10 cases in POAG group, 15 eyes of 13 cases in PACG group,and 6 eyes of 6 cases in SG group.The group of cataract included 12 eyes of 12 cases.And Normal group included 4 eyes of 4 cases.Patients with glaucoma and cataract accepted treatment in the department of Ophthalmology in first hospital affiliated to Chongqing Medical University. Samples of health adult came from donators. Aqueous humor were collected in surgery, a sample of aqueous humor(100-200μl)was manually aspirated into a disposable lml syringe, then immediately transferred to a sterile tube and marked. Put it into a deep freezer at -80℃until assayed. The concentrations of MMP-2 and TIMP-2 in aqueous humor were determined by ELISA kit.The data was analyzed by SPSS 13.0 software.Results: (1) To express the data in mean±standard deviation( x±S) which was consistent with normal distribution. In Cataract group, the concentration of MMP-2 was 13.95±4.40 ng/ml,the concentration of TIMP-2 was 12.84±4.12 ng/ml, and ratio of the two data was 1.03±0.53. In Normal group, the concentration of MMP-2 was 17.81±7.28 ng/ml, the concentration of TIMP-2 was 14.24±5.85 ng/ml, and ratio of the two data was 0.80±0.18. In POAG group, the concentration of MMP-2 was 19.60±5.80 ng/ml, the concentration of TIMP-2 was 46.68±11.64 ng/ml, and ratio of the two data was 2.50±0.71. In PACG group, the concentration of MMP-2 was 23.17±9.22 ng/ml, the concentration of TIMP-2 was 20.88±6.46 ng/ml, and ratio of the two data was 1.02±0.55. To express the data in median±quartile interval (M±Q)which was not consistent with normal distribution. In SG group, the concentration of MMP-2 was 102.60±201.40ng/ml, the concentration of TIMP-2 was 50.87±16.30 ng/ml, and ratio of the two data was 0.49±0.28( x±S).(2) The concentration of MMP-2 in PACG group was significantly higher than Cataract group (P<0.05). The concentration of TIMP-2 in POAG group was significantly higher than other groups(P<0.05). The concentration of MMP-2 in SG group was significantly higher than other groups(P < 0.05).The ratio of the two data in POAG group was significantly higher than other groups(P<0.05).Conclusion: (1) Balance of MMP-2 and TIMP-2 in aqueous humor of patients with primary open-angle glaucoma has been broken. (2) Balance of MMP-2 and TIMP-2 in aqueous humor of patients with primary angle-closure glaucoma has not been broken. (3) The relationship between MMP-2 and TIMP-2 in aqueous humor and patients with secondary glaucoma could be discovered in more further study. (4) Expression of MMP-2 and TIMP-2 in aqueous humor of patients with cortical cataract and nuclear cataract were kept in dynamic equilibrium. In further study,the patients with cortical cataract and nuclear cataract could be regarded as reliable control group.
