ObjectiverThis study was designed to evaluate the relationship between the expression of MMP-2 and TIMP-2 and biological behavior of lung carcinoma.MethodsrThe expression of MMP-2 and TIMP-2 in lung carcinoma and normal lung tissues was determined using immunohistochemical SP technique.Paraffm-embedded specimens from 40 lung carcinoma patients and 20 paracancerous normal lung tissues were examined by Flow Cytometry.Result:Immunohistochemical studies revealed a diffuse granular distribution of MMP-2 and TIMP-2 in the cytoplasm of the carcinoma cells and fibroblast.The expression of MMP-2 and TIMP-2 was stronger in lung carcinoma tissue than in normal lung tissue(pO.Ol). MMP-2 level was found to be significantly higher in lung carcinoma tissues than in paracancerous normal lung tissues examined by Flow Cytometry method (1.52+0.13vs 1.13+0.06, p<0.01).The level of MMP-2 was higher in cases with lymph node metastasis than in those with non-lymph node metastasis(1.69+0.31vsl.41+0.35 p<0.01) The expression ofMMP-2 was negatively correlated to survival time (r=-0.68,p<0.05).TIMP-2 level was found to be significantly higher in lung carcinoma tissues than in paracancerous normal lung tissues examined by FCM (1.24+0.12 vs0.99+0.41 p<0.01).The level of TIMP-2 was higher in cases with lymph node metastasis than in those with non-lymph node metastasis(1.31+0.11vsl.l9+0.09 p<0.01) The expression of TIMP-2 was positively correlated to survival time (r=0.55, p<0.05) . The ratio of MMP-2 /TIMP-2 was higher in cases with lymph node metastasis than in those with non-lymph node metastasis, MMP-2 /TIMP-2 was negatively correlated to survival time (-0.45, p<0.05 ) .Conclusion:1 The expression of MMP-2 and TIMP-2 was observed in lung carcinoma,the positive signals were marked in cytoplasma of carcinoma cells and interstitial cells.The interaction of lung carcinoma cell and interstitial cell is the key to control MMP-2 functional exertion.2 In NSCLC and SCLC , the expression of MMP-2 and TIMP-2 is higher than normal lung tissue ,but the expression of TIMP-2 is lower than that of MMP-2,it proves that the relation of MMP-2 ,TIMP-2 and lung cancer origination from protein level.It indicates that the high expression of MMP-2 is the key in lung cancer origination .3 The expression of MMP-2 in SCLC is higher than that in NSCLC.lt is the reason that SCLC has the ability of highinfiltration and metastasis in clinic.To improve the expression of TIMP-2 will become a biology target to control growth of lung carcinoma.4 The ratio of MMP-2 and TIMP-2 is negatively correlated to lymph node metastasis and prognosis of lung carcinoma and positively correlated to survival time,it indicates that the imbalance of MMP-2 and TIMP-2 is a major reason to result in the cancer origination.lt can act as a reference index in the malignant level of lung carcinoma and prognosis.
