Part I: The establishment , control and observation of the model in cat undergoing cerebral focal ischemia and reperfusion.Objective: To establish a steady , repeatable and perdurable feline model of cerebral focal ischemia-reperfusion and obeserve the model comprehensively by MR, pathology and so on .Materials and Methods: Twenty-two healthy cats were selected into the study, twenty of them were included in operational group and another two were put into sham-operation group. The animals in operational group were rooted on homemade operating-table by anaesthesia . Using sterile microsurgical technique , the left orbit was exenterated and the orbital bone posterior to the the optic foramen were resected with rongeurs to expose the dura overlying the M2 segment of the MCA from the internal carotid artery. The dura and arachnoid were dissected , the M2 segment of the MCA was interrupted by a little medical clip , 2 hours after MCA occlusion the artery were reperfused by undo clip . By using standard methods to nurse and observe animals , neurological function was graded by using the Philips' criteria score. Duiring operation and perfusion of fixed brain we all observed and analysed the M2 segment of MCA . Shortly after reperfusion , to observe the center of thelesion and area around it all animals were undergone functional and conventional MRI and were graded by using the Philips' criteria score at different time. All animals were sacrificed by intracardiac injection of 2.5% formaldehydum plus 1.5% glutaraldehyde and the fixed brain of perfusion was removed by craniectomy , the center of the lesion and area around lesion in specimens were selected based on MR images and stained with HE or TUNEL immunohistochemistry , examined by light microscopy and electro microscopy . Results: after anaesthesia , the animals in operational groups showed diverse symptoms of infarct and neurological deficit ranging from hemiparesis to death . In the area of MAC, the brain tissuse,showed abnormal high intensity in DWI and PWI and abnormal signal in conventional MRI (T1-weighted , T2-weighted imaging and FLAIR).In the area of high intensity in PWI ,we can see a low intensity area, and this area smaller than the area of high intensity in DWI and T2WI at 6 days after reperfusion and the high signal area in T2WI came from the low signal area in PWI at first.The conclusion we reached was evidently different to the study of the graphic-penumbra after cerebral ischemia-reperfusion , namely the high signal area in PWI different from that in DWI .Immunohistochemistry showed a lot of apoptotic cells in the center of the lesion and area around lesion. Electro microscopy examination showed evident swollen in neurons and mitochordria . Some RBC can be seen around the blood vessel , a large number of astrocytes appear in visual field . The severest neurological deficit has occurred between 12 hours to 24 hours after reperfusion .Conclusion: The feline model of cerebral focal ischemia-reperfusion we established has excellent stability , simple operation and reproducibility and was suitable for medical imaging study .
