Objective:Inflammation is one of the important factors in ischemical reperfusion injury. ICAM-1 mediates adherence between leucocyte and VEC, and make leucocyte out of vessel wall into inflammatory brain tissue. In normal, ICAM-1 expresses at a low level, but increases when the stimulations of inflammatory factors exist. The expression of ICAM-1 increases in brain's VEC after focal cerebral ischemia reperfusion. In animal studies, inflammatory cells infiltration can be decreased by suppressing the effect of ICAM-1's stimulating factor or by using anti-ICAM-1 antibody, and animal brain tissue's inflammation can be lighten, infarct volume can be diminished. But there has had no breakthrough in clinical experiment. To administer anti-ICAM-1 antibody via cavitas peritonealis or vein is the traditional way. This study's aim is to approach the feasibility of administration of anti-ICAM-1 antibody via nasal. We hope to expand the sphere in studying ICAM-1.Methods:Thirty male Wistar rats, weighing 200g~250 g, are divided into 5 groups( A, B, C, D, and E,6 every group). Those are sham operation group, cerebral ischemia reperfusion (CIR) group, CIR and high dose antibody group, CIR and middle dose antibody group, CIR and low dose antibody group. CIR model is made by inserting a suture into MCA. Rats are subjected to 2h of ischemia by inserting a suture into the lumen of the ICA and occluding the origin of MCA, followed by 24h of reperfusion. High, middle and low dose anti-ICAM-1 bodies were given to C, D and E group via nasal administration with a sample injector if the animal was lively after having been in ischemia 1h, 15μl each time, ten minutes interval to the next time. Group A is a control group which undergoes a sham-operation. After reperfused the rats were executed. Then the rats were perfused with PBS(4℃, PH=7.4) from left artrium.
