Objective: ( 1 ) To investigate the protein molecular mechanism of tumorheterogeneity of human glioma cell line SWO-38"s two sublines SWOZ1 and SWOZ2. 2 ) To explore a novel methodology for tumor heterogeneity research, we reveal the feasibility of using co-immunoprecipitation in the protein molecular mechanism of tumor heterogeneity.Methods ( 1 ) Mdm2 and p53 expression in glioma cell line SWO-38 and itstwo sublines SWOZ1 and SWOZ2 were detected by immunohistochemical method. 2 ) The expression of Mdm2 and p53 in SWO-38 and its two sublines were measured by Western blot analysis. ( 3 ) The difference at the protein level between Mdm2 and p53 binding proteins expression spectrums of the pair of glioma cell sublines SWOZ1 and SWOZ2 was detected by co-immunoprecipitation method.Results ( 1 ) Mdm2 and p53 expressed in the nuclear of glioma cell lineSWO-38 and its two sublines SWOZ1 and SWOZ2, and the level of three cell lines were similar. (2) Expression of Mdm2 and p53 were observed in SWO-38 and its two sublines, and the level of them were alike. ( 3 )In the Mdm2 immunoprecipitation compound there was an obvious difference between SWOZ1 and SWOZ2, which was in the protein molecular weight of 37 000. And in the p53 immunoprecipitation compound there were three major different bands between SWOZ1 and SWOZ2, which were in the protein molecular weight of 39 000, 38 000 and 37 000.Conclusion ( 1 ) There was a difference in the Mdm2 transmit channelprotein between two sublines and the Mr was 37 000, and there were three different bands in the p53 transmit channel protein between two sublines and the Mr were 39 000, 38 000 and 37 000. It suggest that these different proteins play an important role in tumor heterogeneity of glioma and relate to the different biological characters of those two sub-lines. (2) The different protein in the molecular weight of 37 000 between SWOZ1 and SWOZ2 in the Mdm2 and p53 immunoprecipitation compoundsshould be a same protein, and it should be a intersection between Mdm2 and p53 transmit channel. (3) Co-immunoprecipitation should be useful for the research of protein molecular mechanism of tumor heterogeneity. (4) These two sublines SWOZ1 and SWOZ2 are patent as an ideal model in study of the molecular mechanism of the heterogeneity in glioma.
