Analysis And Correlation Of P53 And Mdm2 Gene Expression In Human Brain Glioma

Objective:To study the express of P53 and MDM2 genes in human in the level of transcription and protein as well as the related genes express study to supply theoretical evidence for clinical diagnosis.Methods:Detect the express of MDM2 and P53 protein in gliomas with LSAB(lakel streptatidin biotin method).Analyze the mRNA expression of MDM2 and P53 genes in gliomas with RT-PCR.Combine imaging operation and the result of pathology to make an approach to statistical analysis.Result:1.The positive rate of MDM2 and P53 protein in glioma was 52%and 41.7%respectively the expression rate of P53 protein in high-level tumour(Ⅲ-Ⅳ)was 26.3%which was distinctly lower than the less-level tumour(Ⅰ-Ⅱ)(69.4%p＜0.05).The expression rate of MDM2 is different in glioma of different levels.It is 68.4%in high-level tumour while 27.6%in less-level tumour(p＜0.05).2.The consistency of two detecting methods about the expression of mRNA in MDM2 is 92.4% 95.2%.The expression of p53 gene is reduced with the increasing of malignant degree,however the expression of MDM2 is improved with the increasing of malignant degree.3.The expression of P53 in glioma is descending with the enlargement of tumour,however,MDM2 is increasing.Conclusion:1.The P53 and MDM2 played different roles in glioma.the former may function as carcinostetic gene in the development of glioma,whereas the later acted a part in carcinoma gene.2.The P53 protein played an important role in the seisure and development of glioma,It was regulated by MDM2 and the regulation was negative fad back,they are negnatively-related. MDM2 played a very important role in the course of glioma.3.There was not only the expression of MDM2 protein but the expression of MDM2 gene was closely connected with the malignant degree and the size of tumour in glioma.The positive rate of MDM2 and P53 protein helps to judge the malignant degree of tumour and forecast the prognosis.
