Objective:Cervical cancer is gynecological malignant tumor with high morbidity rate. In China ,the incidence of this disease has been increasing significantly for a couple of decades. In addition,the age of morbidity is tend to be small. And the mortality of this neoplasia could be greatly reduced by diagnosis and treatment at the earliest stages of the disease. It is of great significance to develop effective detection methods for earlier diagnosis of cervical cancer. Successful treatment of cervical cancer,which depends largely on early detection,is the key to improve survival rate and life qualities of patients vrith cervical cancer. Research has proof that the infection If Human Papillomavirus(HPV) is intimately linked to the cervical cancer. The molecular alternation in cervical cancer consist mainly of inactivating of tumor suppressor genes and activating of oncogene. Inactivating of tumor suppressor genes is a critical event to cervical cancer pathogenesis. To determine the frequency and common deletion region of allelic losses on chromosome and facilitate the isolation of the candidate tumor suppressor genes associated with the cervical cancer. The expression up-regulation of human MutL homologl(hMLH1) gene have something to do with genesis of cervical cancer. hMLH1. Theproliferation cell nuclear autigen (PCNA) had great significance of monitoring the occurrence and development of cervical carcinomas, making the early diagnosis and predicting the prognosis of the patients. In order to provide reference index for pathological diagnosis of cervical cancer, we investgate the role that the infection of HPV, the expression of hMLHl and PCNA have played in the pathogenesis of cancer of the uterine cervix by researching their relationship between them.Methods:Immunchistochemical techniques was used to detect the expression of HPV and hMLHi PCNA of tumor in 82 cases of cervical cancer and 31 cases of chronic cervicitis.Results:The positive expression of HPV hMLH1 and PCNA in 82 cases of human cervical carcinoma were 57.3% 54.9% and 46.3% respectively,while three factor in 31 cases of chronic cervicitis were 64.5% 58.1% and 22.6%,PCNA positive rate in human cervical carcinoma was significantly higher than its of chronic cervicitis(P<0.05).The positive expression of hMLHi and PCNA in human cervical carcinoma of HPV infection positive were 65.2% and 50%,two factor in chronic cervicitis of HPV infection positive were 61% and 22%,while hMLHi positive rate in its of HPV infection positive was significantly higher than its of HPV infection negative(P<0.05).Conclusion These results suggest that HPV could promote expression of PCNA and make much more mispairing of DNA replication which lead to up-regulation expression of products of hMLHi. Detection of PCNA protein and hMLHi gene is helpful for the diagnosis of human cervical carcinoma.
