Bone marrow-derived mesenchymal stem cells (MSCs) are characterized by their abilities of proliferation, self-renewal and multipotent differentiation into various lineages of tissue cells. For decades, intensive attentions have been paid to hematopoietic stem cells that have been achieved great progress. MSCs were thought to play a main role in nutriment and support the growth, proliferation and differentiation of hematopoietic stem cells, so they have been referred to as bone marrow stromal cells. But in recent years, the concept of mesenchymal stem cells has been dramatically widened by the demonstration of their plasticities. Their multipotential has been discovered. MSCs can differentiate into neurons and glia. Further, the special population of non-hematopoietic stem cells is identified as the progenitors of mesoderm-derived tissue cells. These undifferentiated bone marrow mesenchymal stem cells originated from embryonic mesoderm have been successfully cultured in vitro. At present, a suit of consummate method system about culturing and expanding MSCs in vitro has been established. Moreover, MSCs can be induced to differentiate into various lineages of tissue cells, like osteblasts, chondrocytes, myocardial cells, smooth muscle cells, endothelial cells, neuron and astrocytes in the presence of some chemical substances.Cell therapy is considered as a strategy aimed at replacing, repairing or enhancing the biological function of a damaged tissue, organ or system by means of autologous or allogeneic cells transplantation. Potential applied value of MSCs in cell therapy has been cognized gradually along with the study become deeper and deeper. Many aged people are suffering from degenerative diseases, such as Parkinson's disease and Alzheimer's disease, of which the optimal clinic treatment has not achieved up to date. If mesenchymal stem cells can be applied in the cell therapy, these disease disturbed aged people can be conquered. Other diseases, such as nerve system injury, sequelae of operations, ischemic and haemorrhagic diseases, can also be cured by MSCs therapy. But the process of migrating and differentiaion of MSCs in vivo is difficult to be controlled; so mastering differentiation of MSCs in vitro is essential and basal in next clinic therapy. Current therapeusis of nerve system diseases, such as pharmaceutical therapy, interventional therapy and gene therapy, can not achieved content effects to some patients.Some studies have been shown MSCs can differentiate into neuron-like cells or glial-like cells in vivo and vitro. These discoveries provide a new method in clinic therapy. At present, embryonic stem cells already be apllied in clinic, but it is deficient in obtaining, ethic, law and immunological rejection. These problems severely limit clinical utility. To MSCs, they have not these problems. Their good qualities tell their own tale. First, MSCs distribute very abroad. MSCs has been discovered in many tissues and organs derived from mesenchymal tissues; second, they can be obtained from autogeneic bone marrow by simple bone marrow aspiration; third, if they be from autos, cells derived from them can be directly used in transplant therapy, and it is needless to tissue match; others, MSCs is facile in differentiation, though the condition induced differentiation is difficult to control, but MSCs can be easily influenced by the surrounding, they can be induced by many kinds of factors; in addition, mesenchymal stem cells is readly transfected by exogenous gene, and cells decorated by exogenous gene still have the character of highly proliferation and self-renewal. They can be quickly expanded in vitro as cell source in cell therapy. Furthermore, they haven't the ethic and law problem. So MSCs transplant offered a more efficient method in nerve system diseases therapy.Here, on the base of establishing MSCs separating, culturing, purifying and expanding system in vitro, biological characteristics of adult rat bone marrow-derived MSCs and the potential of MSCs differentiating into nerve cells...
