Objective:To establish correspondent animal model in order to observe the dynamic changes of the bile, to do more research about the pathogenesis of pigment stones and techniques of cell culture, moreover, to observe the influence of Qing Re Li Shi herbs to the formation of pigment stones. Methods:1. We established rabbit model, and by means of biochemical method observed the ingredients of bile dynamically including bilirubin, bileacid, Ca +, and so on.2. When the bile excretion was stable, these rabbit were classified into six groups randomly, including Nsgroup, incomplete obstructive group, infective group, LPS group, infective and incomplete obstructive group and LPS and incomplete obstructive group. We also observed the above items of bile during the process of the formation of pigment stones.3. When the bile excretion was stable, we distributed these rabbit into five groups randomly, that is, infective group, LPS group, infective and herb group, LPS and herb group, NS group. And the animals were administrated herbs whose dose is 20 times as many as that of human being. Furthermore, we detected some items of the bile such as bilirubin, bile acid, Ca2+, p-Gase from bacteria and endo-p-Gase. And we also detected the activity of the P-Gase from homogenate of hepatic tissure and Ca2+-Mg2+-ATPase, Na+-K+-ATPase.4. We disgested the rat hepatic cells by using primary lives infusion method, we also observed the influence of the low, medium and high dose herb serum to the activity of Ca2+- Mg2+-ATPase and Na+-K+-ATPase after the incentive of inflammatory factors.Results:1 .The ingredients of model bile had only light changes after 3 days of operation, but there are no significant differences as compared to control group (p>0.05).2.During the formation of pigment stone, infective group, LPS group and incomplete obstructive group didn't have stones. On the contrary, the infective and incomplete group, LPS and incomplete obstructive group had some stones.3.Qi Re Li Shi herbs could decrease the level of UCB and Ca in the bile, increase TEA and lower the activity of p-Gase from both bacteria and hepatic tissue. At the same time, they could also increase the activity of Ca2+- Mg2+-ATPase and Na+-K+-ATPase of the hepatic cellular membrance, so they could resist the formation of pigment stones.4. After the incetive of the inflammatory factors to the herpatic cellular membrance of the rat culture cells, the activity of Ca2+- Mg2+-ATPase and Na+-K+-ATPase were decreased dramatically (p<0.05). Conclusion:his kind of bile model is stable and reliable. We can observe the bile ingredients dynamically, and this model didn't change the size of bile acid pool. We can also get the specimen under a awake condition, therefore, the influence of anesthesia can be excluded. At present, the model can be used to observe many physiological item synchronically. Qi Re Li Shi herbs can lower the occurrence of the pigment stone.
