Part one The Experimental Study on Induction and Differentiation ofthe BMSCs from New Zealand Rabbits ObjectiveTo study the feasibility of induction and differentiation of the bone marrow stromal cells (BMSCs) from New Zealand rabbits in vitro. Materials and methodsUnder the sterile condition, bone marrow stem cells, which came from bone marrow of New Zealand rabbits and were separated by gradient density centrifugation, were cultured, induced and differentiated into neural stem cells and neuronal-like cells with cytokines consisted of retinoic acid (RA, 0.5ug/ml) and glial-derived neurotrophic factor (GDNF, 20ng/ml) and special medium confected in our lab (patent number:02134314.4), and then evaluated by immunocytochemical method. ResultsThe BMSCs of rabbits began to cleavage after 48h , came forth cell clone after 7d, turned into magnus and round cells after 12d, and then changed into neuron-like cells which had long processuses and contacted each other like a net. Cells clone proliferated from BMSCs could express Nestin antigen after cultivated for 7-14 d., while the differentiated cells could express NeuN and NSE antigen after 20d. ConclusionBMSCs of rabbit could proliferate and differentiate into the cells expressing Nestin and NSE antigen. Moreover, as inducer of BMSCs, RA and GDNF played an auxo-role in the process of BMSCs conversion into neural stem cells. Due to its source convenience, the BMSCs could be considerd as an ideal seed cells of the neural stem cells.
