Object To study transdifferentiation of bone marrow-drived mensenchymal stem cells(MSC) into cardiomyocytes after exposured 5-aza and myocardial extracts.Moreover,we have observed the effect of MSC transplantation company with ACEI drug on healing processs after myocardial infarction.Methods 1, after establishing stable culture methods for rat MSC,we have induced MSC with lOumol / L 5-aza at different times. Immunocytochemistry were used to dectect MHC and Troponin-T. 2,we have induced MSC with high level myocardial extracts for 1 week. Immunocytochemistry were used to dectect MHC and Troponin-T. RT-PCR were used to observe expression of a - MHC > ANP and Nkx2.5 genes. 3, in vivo study was performed on rat model of AMI.Surviving rat were randomly divided into different treat groups: MSC transplantation +captopril,captopril(2g/L in drinking water ad libitum) and operation control. MSC labeling with BrdU transplantation were performed in the7day after AMI.On the 21th day after transplantation,some techniques were used for qualitative and quantitative analysis of living MSC,infarct size,thickenss of wall and fibrosis of infarct area.Results After induced by 5-aza,MSC expressed MHC and Troponin-T.,but the positive rate have not showed differentce.After induced by myocardial extracts MSC expressed MHC and Troponin-T, their also expressed a - MHC and Nkx2.5 genes.After transplantation,many cells with BrdU staining positively were observed in infzrct region.Compared with operation control group,Cap raised thickenss of wall. Compared with operation control and Cap groups, MSC transplantation +captopril declined infarct size (40.4 +4.5% vs 24.9 +3.6%, P<0.05) and fibrosis, raisedthickenss of wall (1.65 + 0.41 vs 2.45 + 0.39, P<0.05 ) .Conclusion 1, bone marrow-derived mesenchymal stem cells can differentiate into myocytes when exposured in 5-aza and the differentiation times with 5-aza have no influencee to inducing rate; 2,MSC can transdifferentiatie cardialmyocytes in cardiac microenvironment, and some myocardial soluble macromolrcular substances play a important role in these processs; 4, MSC can survive in infracted region for a long time; 4, on the base of medicine treatment, MSC transplantation can accelerate healing process more effectively after MI.
