Objective: To explore the influence of TMP on vascular smooth muscle cell proliferation and its mechanism.Methods: (1) In vivo test: 40 wistar rats were divided into 4 groups: big dose drug group, small dose drug group, mode group, false operation group. I collagen, IV collagen, PCNA were detected by histochemistry and were detected in FGF, PDGF, c-myc, Bcl-2, BAX by in situ hibridization and TUNEL tests in these paraffin slices . The average light density of every slice were detected by computer image analysis system. The results were analyzed by one-way analysis of variance. And then the results of every group were compared with those of mode group by using Dunnet-t test. (2) In vitro test: Primary cell culture of vascular smooth muscle cell were obtained from human umbilical artery. The cultured VSMC after three more than generations are divided into four groups:high concentration drug group(5000ug/ml), middle concentration drug group(500ug/ml), low concentration drug group(50|j,g/ml), control group. The four groups, after taking drug, were detected in I collagen, IV collagen, PCNA by histochemistry and were detected in BCL-2,BAX by in situ hibridization and TUNEL tests. And then the results of every group were compared with those of control group by using Dunnet-t test.Results: (1) In vivo test: The expression of PCNA in slices of three groups are lower than the mode group. Compared with PCNA results of mode group there is significant difference from the results of false operation group and big dose drug group p<0.05. Compared with PCNA results of mode group there is notsignificant difference from the results of small dose drug group p>0.05. The expression of I collagen, IV collagen, BCL-2, FGF,PDGF, c-myc in slices of three groups are lower than the mode group . Compared with I collagen, IV collagen, FGF, PDGF,c-myc, BCL-2,BAX,and TUNEL test results of mode group there is significant difference from the results of false operation group, small and big dose drug group p<0.05. The expression of BAX in slices of three groups are higher than the mode group .Compared with BAX results of mode group there is significant difference from the results of false operation group, small and big dose drug group ,p<0.05. (2) In vitro test: The expression of PCNA in slices of three concentration drug group are lower than the control group. Compared with the control group, there are markedly significant different from high and middle concentration drug group, p<0.05. There is not significant different from low concentration drug group, p>0.05. The expression of I collagen, IV collagen, BCL-2 in slices of three concentration drug group are lower than the control group .Compared with the control group,there are markedly significant difference, p<0.05. The expression of BAX in slices of three concentration drug group are higher than the control group. Compared with the control group, there are markedly significant difference, p<0.05.Conclusions The results show that TMP can markedly inhibit expression of I, IV collagen,FGF,PDGF in vascular smooth muscle cells , adjust expression of gene relating with VSMC apoptosis and promote VSMC apoptosis.TMP can inhibit VSMC proliferation.
