ObjectiveBy establishing the model of the partial bladder outlet obstruction in male rats,to observe the proliferation, apoptosis of the bladder detrusor cell in different periodsof rat partial bladder outlet obstruction and the change of interstitial collagen fiber.MethodsUsing40male Wistar rats, through the comparison of different methods toestablish rat partial bladder outlet obstruction model, this study utilizes the perineumway ball part of urethra ligation method to establish the model of the partial bladderoutlet obstruction, divided into sham-operated group(10),2weeks of obstruction(15)and4weeks of obstruction(15). We analyze PCNA expression in the bladder detrusorcell by immunohistochemical method. TUNEL method was used to detect the indexof apoptosis cell of bladder detrusor. Applying VG staining to analyze the change ofcollagen fiber in the bladder detrusor.Results1. The selected40male Wistar rats in the experiment, and ultimately survivalonly33, one rat died of deep anesthesia, and two rats died of severe infection, fourrats died of bladder rupture as ligation is too tight.2. The expression of PCNA in the bladder detrusor cell was(17.19±1.37)%、(50.74±3.15)%、(33.58±2.80)%in sham-operated group,2weeks of obstructionand4weeks of obstruction, respectively. By the statistical analysis, there wassignificant difference between2weeks of obstruction and4weeks of obstruction group (P<0.05). The difference between sham-operated group and obstruction groupwas significant (P<0.05).3. The apoptosis index was(20.22±1.70)%、(33.90±1.86)%、(51.18±3.30)%by the TUNEL method in sham-operated group,2weeks of obstruction and4weeksof obstruction, respectively. By the statistical analysis, there was significantdifference between2weeks of obstruction and4weeks of obstruction group (P<0.05).The difference between sham-operated group and obstruction group was significant(P<0.05).4. The proportion of collagen fiber in the bladder detrusor was(7.07±0.67)%、(13.19±1.50)%、(22.98±2.89)%by the method of VG staining in sham-operatedgroup,2weeks of obstruction and4weeks of obstruction, respectively. By thestatistical analysis, there was significant difference between2weeks of obstructionand4weeks of obstruction group (P<0.01). The difference between sham-operatedgroup and obstruction group was significant (P<0.01).ConclusionsPartial bladder outlet obstruction in different obstruction periods, changes ofshape and structure of bladder detrusor may be the bladder detrusor cell proliferation,apoptosis, and changing proportion of interstitial collagen fiber in the combination ofresults.
