Objective To investigate the correlation between the G-A variant at position -30 of the glucokinase gene promoter and type 2 diabetes in Kunming Chinese subjects. Methods Three hundred and eighty-nine unrelated Kunming Han Chinese subjects (279 subjects with type 2 diatetes and 110 normal control subjects ) were studied by polymerase chain reaction-restriction fragment length polymorphism assay. Result Genotype frequency of GG, GA and AA was 63.1%, 31.9% and 5% in type 2 diabetes subjects and 66.4%, 31.8% and 1.8% in normal control subjects. Allele frequency of the A variant was 21.0% in type 2 diabetes subjects and 17.7% in normal control subjects. There was no statistical difference either in genotype or in allele frequency between subjects with type 2 diatetes and normal control subjects. Conclusion: the G-* A variant at position -30 of the GCK gene promoter exists in Kunming Han Chinese and this variant dose not play a major role in the onset of type 2 diabetes; The onset year of type 2 diabetes in AA patients could be earlier than that in GG patients and the body mass index could be smaller than that in GG individuals, this conclusion need to be further studied...
