| Objective To observe expression characteristic of tumor necrosis factor alpha (TNF- α ), interleukin 6 (IL-6) and interleukin 8 (IL-8) in implanted particles of different antigenicity small-ear porcine cancellous bone combined with different amount of small-ear porcine bone morphogenetic protein (sepBMP) and disscuss the immunologic characteristic of combined helerologous bone in implanted processing.Methods The particles of small-ear porcine cancellous bone with differentantigenicity were combined with different amount of sepBMP, implanted in the mouse's muscle pouch , and harvested at the day 5,10,20 after implantation . The specimens were processed to determine the mRNA expression of TNF- α , IL-6 and IL-8 in the area of implanted cancellous bone particles by in situ hybridization technique and observe histological features by HE staining. Resu I ts The expression of TNF- a , IL-6 and IL-8 mRNA was detectable in the area of implantation at the day 5 ,day 10 and day 20 after implantation . The expressions in the fresh cancellous bone carrier groups was markedly higher than the other groups in every group (P<0.05),at the same time, the groups of partially antigen-extracted cancellous bone carrier and antigen-extracted cancellous bone carrier were no significant difference (P>0.05).The expressions of three factors in the groups with no combined with sepBMP were higher than3the groups combined with 2mg sepBMP and 4mg sepBMP (PO.05). The histological observation showed: There was no cartilage cell in the groups with no combined with sepBMP, meanwhile, a few cartilage cells were found in the fresh cancellous bone carrier groups combined with 2mg and 4mg sepBMP ,and at the day 20 was more than the day 10 post-implanted . There were a large amount of cartilage cells , bone-like tissue , primitive marrow cavity formation at the day 10 and 20 postoperatively in the groups of partially antigen-extracted and antigen-extracted cancellous bone carrier combined with 2mg sepBMP and 4mg sepBMP .Cone I us i on The findings indicate that the groups of partially antigen-extracted and antigen-extracted cancellous bone carrier combined with sepBMP occupy high osteogenetic ability with the low evoking immune rejection. TNF- a , IL-6 and IL-8 seem to be important cytokines for the modulation of local cellular immunity in the region of implanted exnogeneic bone. The exogenous sepBMP possesses an inhibitory effect on the expression of TNF- a , IL-6 and IL-8 at the site of implantation . This suggests that sepBMP may be involved in both the low immunity rejection and the induction of osteogenesis in the area of implanted xenogeneic bone. |
