Objective: To study the effect of spleen in acute pancreatitis; and investitute mechanism of spleen in the development of acute pancreatitisMethods: All SD rats were randomly divided into six groups, there are contral class, AP group, AP+tuftsin group, splenectomy+AP group, splenectomy+AP+tuftsin and AP+inhibitor of tuftsin group. AP model was developed in adult rats by retrograde injection of 4% sodium taurocholate in the pancreatic duct, tuftsin or inhibitor group was injected by75ug/kg. At the time of.O. 3. 6. 12h get the pancreas for pathology(HE) and immunohistochemistry(SP), get the serum for detection of TNF and IL-1 by ELISA.Results: Compared contral class,pathological evaluation of all study groups were higher at different times(P<0.01). Pancreatic injury of AP+tuftsin group was more severe than that of AP group at 3 6 12h(P<0.05或 0.01). Pancreatic injury of inhibitor of AP+tuftsin group was milder than that of AP group at 6 12h(P<0.05或 0.01). Pancreatic injury of splenectomy+AP group was more and more severe from 3h to 6h, and had no change from 6h to 12h. Pancreatic injury of splenectomy+AP group was milder than that of AP group at 12h(P<0.01), and had no effect at 3 6h(P>.05). Pancreatic injury of splenectomy+AP+tuftsin group was more and more severe from 3h to 6h to 12h.Compared to AP group, splenectomy+AP+tuftsin group had no effect on Pancreatic injury at different times(P>0.05). Pancreatic injury of splenectomy+AP+tuftsin group was more severe than that of splenectomy+AP group at 6 12h(P<0.05). Microthrombus of splenectomy+AP group was less than that of AP group at 6 12h(P<0.01), and had no effect at 3h(P>.05). Microthrombus splenectomy+AP+tuftsin group was more than AP group at 6 12h(P<0.01),and had no effect on Microthrombus at 3h. Microthrombus of splenectomy +AP+tuftsin group was more than that of splenectomy+AP group at 6 12h(P<0.01). Compared with splenectomy+AP group, Microthrombus of AP+inhibitor of tuftsin group had no change at 3.6 12h (P>0.05).Compared with contral class, TNF and IL-1 of all study groups were higher at different times(P<0.01).
