Objective: To investigate the effects of Tuftsin on inducing apoptosis in hepato-cellular iii order to make research on the principle oftuftsins anti-tumor activity. Methods: 36 mice were randomly divided into six groups: I .sharn-operation; II .total-splenectomy; LII \ IV.total-splenectoiny plus tuftsin injection; V 1/3splenectomy;VI 2/3splenectomy.Three weeks after the operation, comparing the weight of graft carcinoma in these six groups; And peritoneal rnacrophages PM 4, )were drawn out .Using Flow Cytometry to test the apoptosis of hepato-cellular carcinoma in these six groups by PM b. Results: 1 .The apoptosis of peritoneal inacrophages of the total-splenectomy group is lower than that of other groups, while the weight of graft carcinoma is heavier than that of other groups. 2.The apoptosis of sham-operation group is almost the same as the 1/3 splenectorny and 2/3splenectomy and tuftsin injection group. 3 .The apoptosis of hepato-cellular carcinoma induced by the supernatant of PM 4, is similar to that induced by PM 4 4.11w apoptosis of hepato-cellular carcinoma induced directly by tuftsin is more than that of other groups. 5. The apoptosis of hepato-cellular carcinoma is positively correlated with remants of spleen, while negatively correlated with the weight of graft carcinoma. Conclusioiis: 1 .Tuflsin can resist carcinoma by directtly or indirectly inducing carcinoma apoptosis. 2.Tuftsin induces the apoptosis of carcinomaindirectly mainly through the work of supematant of PM 4, 3.Retaining of more than 1/3 of the spleen is able to achieve the aim of resisting carcinoma; whereas, total splenectorny may strikingly reduce the apoptosis of carcinoma and promote the growth of graft carcinoma...
