Objective:To investigate the effect of atorvastatin on aortic expression of Proliferating Cell Nuclear Antigen(PCNA), Tumor Necrosis Factor-α (TNF-α ) and TF in rabbits after establishing the dietary atherosclerotic model, and explore the mechanism. Methods:Eighteen normal male rabbits were randomly and equally divided into three groupsrcontrol group(C group,N=6),fed with basic diet,atherosclerosis group(AS group,N=6),fed with high lipids diet and atorvastatin group,(ATV group,N=6),fed with high lipids diet and treat with atorvastatin. They were fed for 12 weeks, the Aortae were harvested and the concentrations of plasma total cholesterol were measure. We used HE staining and immunohistochemistry staining to measure the protein expression of PCNA and TNF-α .TF expression was analyzed by RT-PCR. Results:1 Qualitative results showed there was no plaque in C group. Plaque both in C group and AS group were aggravated.however, plauqe in AS group was significantly smaller than that in AS group.2 The concentrations of plasma total cholesterol at 12w were 1.30 + 0.12,24.16 + 5.73,12.03 +2.87, respectively,with significant difference among them(P<0.01).3 The number of cells expressing PCNA in 3 group were3.21 +0.45, 289.34+ 21.51,87.53+17.11, respectively.with significant difference among them(P<0.01).4 The number of cells expressing TNF-α were 0,315.47 + 13.61,96.62 + 12.51,respectively,with significant difference among them(P<0.01).5 The relative values of TFmRNA in 3 groups were 0.44 +0.12,1.16 + 0.18,0.74 +0.29, respectively.with significant difference among them (P<0.05). Conclusion:Atorvastatin could remarkably lower plasma total cholesterol. Atorvastatin canreduce excessive expression of PCNA , TNF-α and TF,which may involve the cholesterol-independent effects of atorvastatin.
